芹菜素对雄性大鼠睾丸间质细胞作用机制的研究

目的 研究芹菜素对大鼠睾丸间质细胞(Leydig cells)的作用机制。方法 应用Perooll不连续密度梯度法分离培养大鼠睾丸问质细胞，观察芹菜素对间质细胞睾酮生成的影响。应用MTT方法和流式细胞术观察芹菜素对间质细胞增殖及凋亡的影响。结果 芹菜素作用于大鼠睾丸间质细胞时，以剂量依赖方式使原代培养的大鼠睾丸间质细胞睾酮的分泌量明显减少。MTT及FCM分析显示芹菜素能促进间质细胞凋亡及抑制增殖。结论 芹菜素能不同程度抑制大鼠睾丸间质细胞分泌睾酮并促进睾丸间质细胞凋亡及抑制其增殖。

The Research on the Mechanism of the Effect of Apigenin on the Leydig Cells of Rats

Objective To investigate the mechanism of effects of apigenin (certain phytoestrogens) on the leydig cells of rat testis. Methods Using the technique of Percoll discontinuous density gradients centrifugation, the higher purity of leydig cells were primary cultured. The different concentrations of apigenin were used on leydig cells to investigate the releasing T. The technique of MTT, flow cytometry was used to observe the apoptosis of leydig cells. Results It was indicated that apigenin inhibited T secretion of leydig cells in dose-dependence.FCM and MTT analysis showed that apigenin promoted the apotosis rate of leydig cell and restrained the increment of leydig cells. Conclusions Apigenin could inhibit T secretion of rats leydig cells. Apigenin could also inhibit the reproduction of leydig cells and promoted their apopotis.
