HPLC法测定葛花中鸢尾苷和鸢尾苷元的含量

目的建立测定葛花中鸢尾苷和鸢尾苷元含量的高效液相色谱方法。方法采用美国Agilent-C18柱(4.6 mm×250 mm,5μm),以甲醇-乙腈-水(2∶1∶2)为流动相,流速为1.0 ml/min;检测波长为264 nm,柱温为室温。结果鸢尾苷在2.0～120.0μg/ml呈线性关系,r=0.999 8;平均加样回收率为99.7%,RSD为1.9%;鸢尾苷元在0.8～80.0μg/ml呈线性关系,r=0.998 6;平均加样回收率为99.4%,RSD为1.7%。结论该方法简便易行,结果准确、可靠,重复性好,可用于葛花中鸢尾苷和鸢尾苷元的含量测定。

Determination of tectoridin and tectorigenin in Flos Puerariae lobata by HPLC

Objective To establish an HPLC method for the determination of Tectoridin and Tectorigenin in Flos Puerariae lobata. Method The separation was performed on Agilent -C18 column（4.6 mm × 250 mm,5 um） ,using a mobile phase with methanolacetonitrile-water（2： 1： 2）. Flow rate was 1. 0ml/min,the detection wavelength was set at 264 nm. Column temperature was ambient temperature. Result The linear range of tectoridin was 2.0 - 120.0 ug/ml,r = 0. 999 8. The average recovery was 99.7% ,RSD was 1.9% ;The linear range of tectorigenin was 0.8 -80.0 ug/ml, r = 0. 998 6. The average recovery was 99.4% ,RSD was 1.7 %. Conclusion The method was simple ,accurate,reliable with a good repeatability,which could be used to determinate of tectoridin and tectorigenin in Flos Puerariae lobata.
