骨髓间充质干细胞移植在急性肝损伤大鼠肝脏内定植能力的实验研究

摘    要:目的:观察大鼠骨髓间充质干细胞(BMSCs)在急性肝损伤大鼠肝脏内移植情况.方法:首先行大鼠骨髓间充质干细胞提取、分离和培养,传代扩增后用5-溴脱氧尿嘧啶核苷(BrdU)标记,用D-氨基半乳糖(1.5g/Kg)腹腔注射制备大鼠急性肝损伤模型,试验分三组(A、B、C组),经阴囊上静脉向肝损伤组(A组)及肝脏正常(C组)的大鼠移植标记BrdU的BMSCs 1~1.5x106,同时向肝损伤组(B组)的大鼠移植等量的生理盐水.2周后观察大鼠的存活率及肝功能恢复情况,并通过免疫组织化学方法检测大鼠肝脏中BrdU 细胞数量及分布.结果:移植2周A组、B组动物存活率、肝功能恢复情况无明显差异,肝损伤组(A组)及肝脏正常组(C组)大鼠肝脏均可检测到BrdU 细胞分布,A组与C组相比,A组BrdU 细胞数较多,分布更广,差异有统计学意义(P<0.05).结论:骨髓间充质干细胞可以在受损的肝脏及正常肝脏定植,定植的数量可能和肝脏是否受损伤相关.

Abstract:Objective:To observe the homing capacity of bone mesenchymal stem cells(BMSCs) to rat with acute hepatic failure(AHF). Methods:BMSCs were isolated from rat bone marrow with the method of density gradient centrifugation and stick-to-wall screening,and the surface markers were identified with flow cytometry after amplification in vitro. BMSCs were labeled with Bromodeoxyuridine(BrdU). AHF animals were induced by D-galactosamine(D-gal). Transplantation of labeled BMSCs(1~1.5×106) were performed on the AHF rats(group A)/normal rats(group C),another AHF group(group B) were injected with sodium chloride without BMSCs as a control to group A. Then the mortality of the rats and liver function recovery status were observed. Two weeks after cells transplantation, BMSCs traced with BrdU were detected in the livers by immunohistochemistry. Results:The mortality and status of liver function were not significantly different between A and B group. Both A and C group could find BrdU+ cells, and the number and the disposition of BrdU+ cells in A group was much more than C group-s,showing significant difference. Conclusion:BMSCs have capacity of homing to the liver of rat in normal or in injury . But the number of BMSCs homing to liver might be related to the extent of liver injury.
