溶剂回流法提取香椿叶总黄酮的工艺研究

摘    要:目的:研究香椿叶总黄酮的提取工艺。方法:以料液比、提取温度、提取时间、乙醇体积分数和提取次数为考察因素,选用溶剂回流法进行总黄酮提取;以总黄酮的提取量和提取物以黄酮计的清除2,2-二苯代苦味酰基苯肼基(DPPH)的半数有效值(IC50)为评价指标进行正交试验。结果:最佳提取工艺条件为料液比1:9、温度70℃、提取时间90min、乙醇浓度70%、提取4次。香椿叶总黄酮提取量为20.1545mg.g-1,IC50=24.595g.g-1。结论:溶剂回流法提取香椿叶总黄酮的工艺可行。

Abstract:OBJECTIVE: To study the optimum extraction condition of total flavonoids from Chinese Toon leaves by solvent backflow method .METHODS: The total flavonoids was extracted from Chinese Toon leaves by solvent backflow method taking the percent of ethanol volume, the ratio of material to liquid, time,temperature and times of extraction as factors.Orthogonal experiment was performed with extractive yield of total flavonoids and IC50 of scavenging DPPH free radicals of total flavonoids as indexes.RESULTS: The optimum technical extractive conditions of total flavonoids from Chinese Toon leaves were as follows:the extraction temperature was 70 ℃ ;the concentration of ethanol was 70 %, the ratio of solid to liquid was 1 : 9;the extracting lasted for 90 min each time for a total of 4 times.Under this condition, the yield of total flavonoids extracted from Chinese Toon leaves was 20.154 5 mg · g ^-1 and the IC50 of scavenging DPPH free radicals of extracts was 24. 595 g · g^ -1. CONCLUSION: The solvent backflow extraction method is feasible for the extraction of total flavonoids from Chinese Toon leaves.
