| 摘 要： | 目的评价Vero/Slam细胞对麻疹病毒的敏感性,以及麻疹病毒在Vero/Slam细胞上的生长特点。方法组织细胞培养法分离病毒,将标本接种到长满单层的Vero/Slam细胞上,盲传3代,观察细胞病变(CPE)。将分离到的麻疹病毒通过逆转录-聚合酶链反应(RT-PCR),扩增出核蛋白(N)基因碳末端450个核苷酸片段,对其产物测定基因序列以定型。结果Vero/Slam细胞对麻疹病毒高度敏感,其CPE特点是典型的细胞融合性病变。此次分离到的麻疹病毒为H1基因型,和中国的本土优势流行株相符。结论Vero/Slam细胞对麻疹病毒敏感,可在麻疹病毒分离中选用。

| Abstract: | Objective To study the sensitivity of Vero/Slam cell to measles virus(MV) and characteristics of MV in Vero/Slam cell.Methods Histiocyte culture.The samples were inoculated in the single layer Vero/Slam cells,up to 3 blind passages,and observed the MV CPE.The MV from these samples were isolated using Vero/Slam cell line,450 nucleotides of COOH-terminal of nudeoprotein genes were amplified and sequence analyzed by RT-PCR and sequence analysis software.Results Vero/Slam cell line is highly sensitive to measles virus.The cytopathic effect(PE) characteristics of measles virus in Vero/Slam cell was cell fusion.The isolated wild-type measles virus is H_1,the same geno-type as Chinese native prevailing measles virus.Conclusion Vero/Slam cell line is sensitive,it can be used in isolation of measles virus. |
