一氧化氮合酶与脑缺血亚急性期神经损伤的关系

摘    要:目的 探讨大鼠脑缺血亚急性期脑力内三型一氧化氮合酶(NOS)的表达及其与经济损伤的关系。方法 采用大鼠大脑中动脉缺血模型,用免疫组织化学方法检测局灶性脑缺血24h三型NOS在脑内的表达。结果 局灶性脑缺血24h,缺血中心区及部分边缘区内可见大量诱导型一氧化氮合酶(iNOS)阳性的胶质细胞浸润。表达神经元型一氧化氮合酶(nNOS)及内皮型一氧化氮合酶(eNOS)的神经细胞数减少。结论 脑缺血亚急性期胶质细胞iNOS表达上调与迟发性神经元损伤有密切关系。在此期给予选择性iNOS抑制剂可以减少缺血性损害。

Abstract:Objective\ To study the expression of three isoforms of nitric oxide synthase(NOS) and it's relation with neuronal damage at the subacute stage of focal cerebral ischemia in rat.\ Methods\ In a model of middle cerebral artery occlusion(MCAO) through monofilament nylon suture.\ Three kinds of NOS polyclone antibody\|SP immunochemisty technique were used to detected the expression of neuronal NOS(nNOS), inducible NOS(iNOS) and endothelial NOS(eNOS) after 24 h focal cerebral ischemia.\ \{Results\}\ 24 h after permanent focal cerebral ischemia, plenty of iNOS immunoreactivite glial cell were seen in the core and the periphery of the ischemic lesion, but the expression of nNOS and eNOS were downregulated in the infarct region.\ Conclusion\ The upregulated expression of iNOS in glial cells at the subacute stage of focal cerebral ischemia may be related to delayed neuronal cell death.\ It suggests that the use of selective iNOS inhibitors could be beneficial to reduce the neuronal ischemic damage.
