Comparison of Two Methods of Site - specific Mutagenesis

Two methods of site-specific mutagenesis were used to delete 192 bp for constructing a new expression vector and a comparison was carried out. The designed deletion was completed by means of the U-containing system and phage M13 system, with efficiency of 6% and 1% respectively. The conclusion is that the U=containing system produces high mutants, although it needs special agents, bacterial strains and more complicated manipulations, whereas the phage M13 system is easy to perfom, and needs no special agents or bacterial strains. The time taken for the later method is shorter but the mutation rate is rather low.
