摘 要: | ![]() 目的 研究miR-137 过表达对人胃癌MKN-45 细胞迁移侵袭的影响. 方法 采用RT-PCR法检测5 株胃癌细胞和人胃黏膜上皮细胞( GES )中 miR-137 的表达. 构建miR-137慢病毒,并转染MKN-45细胞. 通过划痕实验、细胞侵袭小室检测和Transwell侵袭实验以分析过表达miR-137对MKN-45细胞迁移侵袭的影响. 结果 MKN-45 细胞中miR-137的表达丰度明显低于在 GES、MKN-74、AGS、SGC-7901和BGC-823细胞,差异有统计学意义(P<0. 05). 同时成功构建micro up病毒感染的细胞组. 划痕实验结果显示,与空白对照组( CON组)和阴性对照组( NC组)比较, micro up组24、48 h后平均迁移率差异有统计学意义( P<0. 05 );细胞侵袭小室检测结果显示,与 CON组、NC组比较, micro up组的平均迁移率差异有统计学意义( P<0. 01 );Transwell侵袭实验结果显示,与CON组、NC组比较,micro up组平均迁移率差异有统计学意义( P<0. 01 ). 结论 miR-137过表达能明显抑制人胃癌MKN-45细胞迁移侵袭能力,有可能成为胃癌治疗的新靶点.

Effect of microRNA-137 on migration and invasion of MKN-45 cells of human gastric cancer |

Abstract: | ![]() Objective To study the effectiveness of over expression of miR-137 on migration and invasion of human gastric cancer MKN-45 cells. Methods RT-PCR technique was explored to detect the expression of miR-137 of 5 strain of gastric cancer cell and human gastric epithelium. Constructed lentivirus vector of overexpressing miR-137. Scratch test, cell invasion chamber test and Transwell invasive test were used to analyze the effectiveness of overex-pression of miR-137 on migration and invasion of human gastric cancer MKN-45 cells. Results The degree of ex-pression of miR-137 in MKN-45 was obviously lower than those in GES, MKN-74, AGS, SGC-7901 and BGC-823 cells and the difference was significant(P<0. 05). The lentivirus vector of micro up miR-137 infected cells group was successfully built. Scratch test result showed that the average migration rates after 24, 48 h of micro up group compared with CON group and NC group, the difference was significant (P<0. 05). The results of cell invasion chamber test and Transwell invasive test all showed that the average metastatic rates of micro up group compared with CON group and NC group, the difference was significant (P<0. 01). Conclusion Micro up miR-137 could inhibit distinctively the ability of migration and invasion of human MKN-45 cell strain and might be a novel target point for treatment of gastric cancer. |
