摘 要: | 目的:探讨辅助生殖技术中巨型卵母细胞的染色体组成以及对于胚胎生长发育的影响。方法:收集40例患者的44枚巨卵细胞,分析了巨卵的成熟度、受精和胚胎发育情况,并采用荧光原位杂交(fluorescence in situ hybridization,FISH)技术分别检测了巨卵及其胚胎的X、13和14号染色体组成。结果:巨卵的发生率约为0.29%,巨卵细胞的平均直径显著大于普通卵母细胞(P<0.05)。获得巨卵的40例患者的平均获卵数明显高于未产生巨卵的患者(P<0.05)。10个巨卵细胞的细胞遗传学检测显示其均为二倍体,对5个2PN巨卵胚胎的13个卵裂球行FISH检测的结果显示,5个胚胎为三倍体或者嵌合体。结论:巨卵受精可能是形成母源性三倍体的原因之一。鉴于其染色体数目异常,对于巨卵发育而来的胚胎可能并不适合用于移植。

Effect of gaint oocytes on embryonic development in assisted reproductive technology |

Abstract: | Objective:To explore the chromosomal composition of gaint oocytes in assisted reproductive technology and its impact on embryonic growth and development.Methods:Forty-four gaint oocytes from 40 patients were collected,the degree of maturity,fertilization,and embryonic development of gaint oocytes were analyzed,fluorescence in situ hybridization(FISH) technique was used to detect the compositions of X,13,and 14 chromosomes of gaint oocytes and embryos.Results:The incidence rate of gaint oocytes was 0.29%,the mean diameter of gaint oocytes was statistically significantly larger than that of general oocytes(P<0.05).The mean number of retrieved oocytes in 40 patients with gaint oocytes was statistically significantly higher than that in patients without gaint oocytes(P<0.05).The results of cytogenetic detection showed that ten gaint oocytes were diploid,the results of FISH detection about 13 blastomere of five 2PN gaint oocytes showed that five embryos were triploid or chimera.Conclusion:Fertilization of giant oocytes may be one of causes of maternal triploidy.In consideration of abnormal chromosome number,the embryos derived from gaint oocytes may be unsuitable for embryo transfer. |
