人结肠癌ccl227细胞系肿瘤干细胞生长特性研究

目的：研究体外培养条件下人结肠癌ccl227细胞系肿瘤干细胞的生长特性。方法：用荧光染料Hoechst33342染色培养在SSM和SFM中的ccl227细胞，荧光显微镜观察SP细胞。用6孔板接种的方法绘制ccl227细胞的生长曲线。显微镜观察肿瘤球的生长。结果：在两种培养条件下均观察到ccl227细胞系中存在浅染和拒染的SP细胞。生长曲线显示在两种培养条件下，ccl227细胞的繁殖能力不同。在玻璃培养瓶和低吸附6孔板中均可以培育出肿瘤球，但形态有些不同。结论：SSM和SFM培养条件下，ccl227细胞中均存在SP细胞；在SFM中，ccl227细胞增殖缓慢；SFM可培育出富含肿瘤干细胞的肿瘤球。在低吸附条件下，更易形成悬浮的肿瘤球。

Study of cancer stem cell characteristic of human colon cell line ccl227

Objective：To study the characteristic of tumor stern cell of human colon cancer line ccl227 under different condition. Metbods：Ccl227 cell cultured in SSM and SFM were viewed by fluorescence microscopy after being stained with Hoechst33342. The growth curve of ccl227 cell was made by cell counting. The growth of tumor sphere was viewed by microscopy. Resulsts ： Under - stained SP cells were seen in SSM and SFM. Some of them were not stained. The productivity of ccl227 cell cultured in SSM and SFM was different. The tumor sphere was viewed in SFM. Conclusion：There are SP cells in ccl227 cell cultured in SSM and SFM. The growth of ccl227 is slowly in SFM. Tumor sphere can be generated in SFM. Under low adsorbing, tumor sphere can generate more easily.
