大黄酚固体分散体对镉染毒大鼠氧化应激影响

摘    要:
目的 观察大黄酚固体分散体对镉染毒大鼠氧化损伤的影响.方法 以聚乙二醇6 000为载体制备大黄酚固体分散体,差示扫描量热法(DSC)和X射线衍射法对固体分散体进行分析.SD大鼠40只,雌雄各半,随机分为5组,分别为正常对照组、镉染毒组、镉加大黄酚组、镉加大黄酚固体分散体低剂量组和镉加大黄酚固体分散体高剂量组.正常对照组灌胃纯净水,其余4组给予20 mg/kg氯化镉隔日灌胃染毒,共染毒4次,染毒6h后给予大黄酚或大黄酚固体分散体干预.末次给药后,测定大鼠血清、脑和肾中超氧化物歧化酶(SOD)活力及丙二醛(MDA)水平,测定大鼠血清谷胱甘肽过氧化物酶(GSH-Px)、丙氨酸转氨酶(ALT)和天冬氨酸转氨酶(AST)活力.结果 DSC和X射线衍射分析表明,大黄酚以无定形形式存在于固体分散体中.大黄酚固体分散体可降低镉染毒大鼠血清、脑组织和肾中MDA水平(P<0.05),增加血清、脑组织和肾中SOD和GSH-Px活力(P<0.05),降低血清中ALT、AST活力(P<0.05).结论 大黄酚固体分散体可降低镉对大鼠造成的肝损伤、脑损伤和氧化损伤.
