黄连干预心肌缺血损伤的体内外实验研究

摘    要:目的利用体内、体外实验模型研究单味黄连对心肌(细胞)缺血、缺氧的保护作用及机制。方法结扎左冠状动脉复制大鼠心肌缺血模型,灌胃给予黄连粗提物61.27 mg/(kg.d),每日1次,7 d后测定心电图、血清酶学指标;建立缺氧复氧心肌细胞模型,给予黄连粗提物(25μg/mL),测定细胞培养液中乳酸脱氢酶的漏出和细胞活力。结果黄连能对抗心肌缺血引起的Ⅱ导联心电图ST-T段的抬高(P0.05),降低血清肌酸激酶、乳酸脱氢酶、丙二醛(P0.05),升高超氧化物歧化酶(P0.05);减少缺氧复氧心肌细胞乳酸脱氢酶的漏出(P0.05),而不影响细胞相对活力。结论黄连具有保护心肌(细胞)作用,其机制是通过改善心肌(细胞)缺血、氧化损伤状态而实现。

Protective Effects of Rhizoma Coptidis on Acute Myocardial Ischemia Injured Cardiomyocytes in Vivo and in Vitro

Abstract:Objective To investigate the cardioprotective effects and the mechanism of Rhizoma Coptidis (RC) on acute myocardial ischemia in vivo and in vitro. Method Male SD rats were randomized into four groups: blank control group, model group, RC group ig, 61.27 mg/(kg · d)], and diltiazem hydrochloride group ig, 37.8 mg/(kg · d)]. After 7 days administration of the crude extracted RC, the animal model of acute ischemic myocardial injury was estabhshed by hgaturing the left anterior descending coronary artery (LAD) in rat hearts. Also, a model of hypoxia/reoxygenation injury in cultured neonatal rat cardiomyocytes was used. The electrocardiogram (ECG), levels of creatine kinase (CK), lactate dehydrogenase (LDH} and malonaldehyde (MDA), acticity of superoxide dismutase (SOD) in serum were measured. Result Compared with the model group, RC significantly decreased the level of ST-T (P〈0.05) in ECG, and the levels of CK, LDH and MDA (P〈0.05) in serum, which were increased by acute myocardial ischemia. The activity of SOD was improved by RC treatment. RC markedly decreased the level of LDH (P〈0.05) caused by hypoxia/reoxygenation injury in cultured neonatal rat cardiomyocytes. Conclusion The crude extract of RC had a protective effect on cardiomyocytes in rat. The mechanism of RC might be through improving the oxidative damage of acute myocardial ischemia.
