降糖消脂片对糖尿病大鼠视网膜血管的保护作用

目的研究降糖消脂片对糖尿病大鼠视网膜病变的作用。方法 SD大鼠以链脲佐菌素50 mg/kg腹腔注射造模。3 d后测定血糖,筛选成模大鼠。将造模成功大鼠随机分为模型组、空白组、阳性药组及降糖消脂片低、中、高剂量组,每组12只。降糖消脂片低、中、高剂量组分别给予降糖消脂片2、4、8 g/kg体质量,阳性药组给予二甲双胍0.3 g/kg体质量,模型组和空白组给予等体积蒸馏水。给药8周后处死大鼠,分离视网膜,进行PAS染色,观察各组血管形态,计数非细胞血管、血管密度、血管条索数、血管周细胞与内皮细胞数量。结果降糖消脂片各剂量组给药后能明显保持视网膜血管网的生理结构,明显减少视网膜非细胞血管数、降低血管密度、减少血管条索数、增高周细胞与内皮细胞比值。结论降糖消脂片对糖尿病视网膜血管具有明显的保护作用。

Protective Effect of Jiangtang Xiaozhi Tablets on Retina Capillary in STZ-Induced Rat Diabetic Retinopathy

Objective To observe effects of Jiangtang Xiaozhi tablets on streptozocin(STZ) induced rat diabetic retinopathy.Methods SD rats were intraperitoneally injected STZ at dose of 50 mg/kg.The levels of blood glucose in the rats were measured after 3 days for screening model animals.Thereafter,rats were divided into model group,control group,positive drug group and Jiangtang Xiaozhi tablets low-,medium-,high-dose group,12 rats for each group.Jiangtang Xiaozhi tablets group were continuously given extract 2,4,8 g/kg by intragastric administration for 8 weeks.Positive drug group was given metformin 0.3 g/kg.Model group and control group were given distilled water.Rats were killed and retina were isolated for PAS stain.Morphology of retina vascular was observed,and acellular capillaries,microvessel density pericyte numbers and endothelial cell numbers were counted on trypsin digests.Results Jiangtang Xiaozhi tablets administration could protect the morphology of retina vascular structure,significantly reduce the number of acellular capillaries,blood vessel density,and the number of vascular cord.Meanwhile,it obviously increased the ratio of pericyte and endothelial cell.Conclusion Jiangtang Xiaozhi tablets have significant protective effect of retina capillary in STZ-induced rat diabetic retinopathy.
