莫索尼定滴眼液点眼对家兔眼压及血压的影响

摘    要:目的探讨不同剂量莫索尼定(MOX)滴眼液点眼对眼压、瞳孔、血压和心率的影响。方法 32只家兔全身麻醉后行双眼前房穿刺并连接于生理记录仪描记眼压,行单侧颈动脉插管并连接于生理记录仪描记血压、心率波动。分别将质量分数0.05%、0.1%和0.2%的MOX单侧点眼,同步观察点眼后8h内双侧眼压、瞳孔、血压和心率的变化。结果 0.05%、0.1%、0.2%MOX组点眼后实验眼最大眼压下降幅度分别为3.01mmHg(18.31%)、3.70mmHg(22.13%)和4.46mmHg(25.60%),降眼压持续时间分别为6h、8h和8h以上。对侧眼眼压下降持续时间较短,最大眼压下降幅度分别为2.64mmHg(15.46%)、3.10mmHg(18.82%)和2.62mmHg(16.48%)。3组MOX滴眼液点眼均可使实验眼瞳孔散大,最大散瞳幅度分别为2.6、3.2、3.9mm。MOX滴眼液点眼对心率影响轻微,0.05%MOX组和0.1%MOX组点眼不影响实验动物的血压,0.2%MOX组可使实验动物舒张压下降,最大下降值为8.85mmHg(9.93%)。结论 MOX滴眼液点眼对家兔有降眼压作用,中低剂量的MOX对实验动物的血压、心率等无明显影响。

Abstract:Background Moxonidine is an agonist of α2 receptor.It has been used to the clinical treatment of glaucoma.Therefore,the effects of moxonidine on ocular tissue and blood pressure and heart rate is also interested research topic.Objective This study was to evaluate the effects of different concentrations of moxonidine on intraocular pressure (IOP),pupil diameter (PD),heart rate (HR) and blood pressure (BP) simultaneously after topical administration.Methods 0.05%,0.1%,0.2% moxonidine were topically used in the lateral eyes of healthy white rabbits in 0.05% moxonidine group (8 rabbits),0.1% moxonidine group (8 rabbits),0.2% moxonidine group (8 rabbits) respectively and sterilized distilled water was topically used in the control group(8 rabbits).IOP of the rabbits was measured by puncture of anterior chamber with 41/2 gauge needle connecting physiological recorder,and the diameter of pupil was measured using ruler before usage of the drug and 0.5,1,2,3,4,5,6,7,8 hours after application of the drug.BP and HR of the rabbits were detected with carotid artery catheter recorder.Results Moxonidine caused dose-dependent decreases in IOP.The maximum decreases in IOP were 3.01 mmHg (18.31%).3.70 mmHg(22.13%),4.46 mmHg(25.60%) and ocular hypotension lasted for 6,8 and over 8 hours in ipsilateral eyes at the three doses of moxonidine respectively.In the contralateral eyes the duration of ocular hypotension were shorter,the maximum decreases in IOP were 2.64 mmHg(15.46%),3.10 mmHg (18.82%)and 2.62 mmHg (16.48%) respectively.All doses of MOX caused significant increases in PD (the maximum increases were 2.6 mm,3.2 mm and 3.9 mm) in ipsilateral eyes and did not affect HR.The largest dose of MOX reduced diastolic blood pressure.The maximum decreases was 8.85 mmHg(9.93%).The other two doses of MOX didn’t affect BP.Conclusion Moxonidine produces ocular hypotension and small dose and middle dose of moxonidine has no influence on BP and HR when applied topically in rabbit eye.
