Ang-1和Ang-2在大肠癌中的表达和临床病理特征的关系

目的:研究血管生成素1,2(Ang-1,Ang-2)蛋白在大肠癌组织中的表达及其与大肠癌临床病理特征的关系。方法:应用免疫组化法检测50例大肠癌患者及正常人大肠黏膜组织中Ang-1、Ang-2蛋白表达水平。结果:大肠癌组织中Ang-2蛋白表达阳性率为78%(39/50),明显高于正常大肠黏膜组织的33.3%(5/15)(P<0.01),大肠癌组织Ang-1蛋白表达阳性率为46%(23/50),大肠黏膜正常组织Ang-1蛋白表达阳性率为66.7%(10/15)。Ang-1蛋白在正常组织和大肠癌组织中均有表达,两者差异无统计学意义(P>0.05),Ang-2蛋白表达水平与患者性别、年龄及癌的体积、癌分化程度、淋巴结转移无关(P>0.05),Dukes分期D期Ang-2蛋白表达水平明显高于A期(P<0.05)。有血道转移的比无血道转移的Ang-2表达水平高(P<0.05)。肠壁浸润越深,Ang-2蛋白表达水平越高(P<0.05)。结论:Ang-2蛋白表达水平与癌的肠壁浸润深度、临床分期及大肠癌血道转移有关,提示Ang-2蛋白高表达可能促进大肠癌的生长及转移;Ang-1蛋白表达与癌的发生无关。

Expression ofangiopoietin-1,2 and relationship of clinical parameters in colorectal carcinoma

Objective:To study the expression of Angiopoietin-1,2 (Ang-1 and Ang-2) protein in colorectal carcinoma and evaluate the relationship between Ang-1,2 protein level and clinical parameters. Methods:Expression of Ang-1,2 was examined in 50 patients with colorectal carcinoma and normal tissues with immunohistochemical technique. Results:The positive rate of Ang-2 protein was significantly higher in colorectal carcinoma than in normal tissues(78% vs 33.3 %, P< 0. 01) . The positive rate of Ang-1 protein was 46 %(23/ 50)in colorectal carcinoma .The positive rate of Ang-1 protein was 66.7 %(23/ 50)in normal tissues. Ang-1 protein expressed both in colorectal carcinoma and normal tissues ,there was no obvious difference (P>0.05). Neither patient's gender nor age correlated with the expression level of Ang-2 (both P>0.05). No correlations were found between the expression level of Ang-2 and the size or histological differentiation of colorectal carcinoma (P>0.05). The expression level of Ang-2 in colorectal carcinoma with lymph node metastasis was not different from that without lymph node metastasis(P>0.05). The Ang-2 positive rate was lower in colorectal carcinoma of Dukes'A stage than that of Dukes'D stage (P<0.05),and was higher in colorectal carcinoma with metastasis from blood vessels than that without metastasis from blood vessels(P<0.05).The deeper extent of infiltration, the higher expression of Ang-2 protein. Conclusion:The expression level of Ang-2 protein may be related to the shape of colorectal carcinoma , invasion ability, Dukes' staging in colorectal carcinoma, and protein may play an important role in the progression and metastasis of colorectal carcinoma. It is suggested that high expression of Ang-2 rectal carcinoma. No correlations are found between the expression level of Ang-1 and the progression of coloblood vessel metastasis .
