四种HIV分子流行病学分析方法的比较研究

摘    要:
目的 比较艾滋病病毒1型(HIV-1)血清学法(PEIA)、异源双链泳动法(HMA)、基因序列分析法(GSA)和基因芯片分析法四种亚型分析方法的准确性及特异性,评价其优劣及对于结果判定的影响。方法 分别用上述四种方法对25份经初筛及确认试验认定为HIV-1阳性的血清标本进行亚型分析。结果 PEIA亚型分析结果:B亚型12份,C亚型7份,E亚型6份;HMA亚型分析结果:B亚型13份,C亚型8份,E亚型4份;GSA亚型分析结果:B亚型13份,C亚型9份,E亚型3份;基因芯片法亚型分析结果:B亚型13份,C亚型9份,E亚型3份。结论 HIV-1四种亚型分析方法敏感性和特异性均较好,其中基因芯片分析法与目前通用的GSA所得结果相同,且操作简单,省时价廉,适用于大规模的标本分析,此法值得推广应用。

Abstract:
Objective To evaluate the sensitivity and specificity of four HIV-1 molecular-epidemiological analytic methods Methods The subtypes of 25 HIV-1 serum specimens were analyzed,using peptide-binding enzyme immunoassay(PEIA),heteroduplex mobility assay(HMA),gene sequences analysis(GSA)and genechip assay Results For PEIA subtype B,subtype C and subtype E were identified in 12,7 and 6 specimens,respectively;for HMA subtype B,subtype C and subtype E were identified in 13,8 and 4 specimens,respectively;for GSA subtype B,subtype C and subtype E were identified in 13,9 and 3 specimens,respectively;and finally for genechip assay subtype B,subtype C,and subtype E were identified in 13,9 and 3 specimens,respectively Conclusions PEIA,HMA,GSA and Genechip were all sensitive and specific in determining HIV-1 subtypes The results of genechip were the same as those of GSA,and genechip assay can be used widely due to its low price and simplicity
