糖基化终产物对人脐静脉内皮细胞血管内皮生长因子mRNA及蛋白表达的影响

摘    要:
目的研究糖基化终产物(AGEs)对人脐静脉山皮细胞血管内皮生长凼子(VEGF)mRNA及蛋白表达的影响,探讨AGEs在糖尿病动脉粥样硬化中的作用。方法将人脐静脉内皮细胞株ECV304用BSA及小同浓度的AGEs孵育24h及400mg/L的AGEs孵育0、12、24及36h,采用原位杂交及Westem blot方法检测VEGFmRNA及蛋白的表达。结果人脐静脉内皮细胞在100、200及400mg/LAGEs孵育24h后,VEGFmRNA及蛋白表达均显著高于BSA对照组(P〈0.05),在用400mg/LAGEs分别孵育12.24及36h后,各组内皮细胞VEGF mRNA及蛋白表达量也明显高于0h对照组(P〈O.05)。结论AGEs可增加人脐静脉内皮细胞VEGF mRNA及蛋白的表达,且与浓度和时间呈正相关。

Abstract:
Objective To study the effects of advanced glycosylation end products(AGEs) on mRNA and protein expression of vascular endothelial growth factor(VEGF) in human umbilical vein endothelial cells(ECV304) and to explore the function of AGEs and VEGF in atherosclerosis.Methods HUVECs(ECV304) were cultured with AGEs at different concentrations and BSA for 24 h and at a concentration of 400 mg/L for 0,12,24 and 36 h.In situ hybridization and Western blot were used to detect the levels of mRNA and protein expression of VEGF.Results The expression of VEGF mRNA and protein in HUVECs to AGEs at different concentrations(100,200,400 mg/L) for 24 h were higher than that in BSA control group(P<0.05),the expression at a concentration of 400 mg/L for 12,24 and 36 h were higher than that in 0 h control group(P<0.05).Conclusions AGEs could increase the expression of VEGF mRNA and protein in a time and dose dependent manner in HUVECs.
