苯丙氨酸对胚鼠大脑皮层神经元Rho GTPases基因表达的影响

摘    要:目的研究苯丙氨酸对大脑皮层神经元RhoGTPases(Racl、Cdc42、RhoA)mRNA体外表达的影响。方法在体外培养3d的原代培养胚鼠大脑皮层神经元中,加入高浓度苯丙氨酸诱导18h,用实时荧光定量PCR方法,检测苯丙氨酸对胚鼠大脑皮层神经元Rac1、Cdc42、RhoA基因表达的影响。结果胚鼠大脑皮层神经元经苯丙氨酸诱导后,Rac1、Cdcd2、RhoAmRNA表达分别是对照组的37.0%、29.8%、30.0%(P<0.01)。结论高浓度苯丙氨酸通过影响RhoGTPases基因表达,进而干扰神经元突起的生长和突触的连接,可能是苯丙酮尿症导致患者智力落后机制之一。

Abstract:Objective To determine the genes expression of Rho GTPases including Rac1 , Cdc42 , RhoA in the embryonic rat cerebral cortical neurons induced by phenylalanine. Methods Primary embryonal rat cerebral cortical neurons were cultured for 3 d and the cells induced by phenylalanine (0.9 mmol/L) for 18 h. Real time PCR was used to determine the influence of phenylalanine on the genes expression of Rho GTPases. Results In our experimental condition, the genes expression level of Racl , Cdc42 , RhoA in phenylalanine group were significantly decreased (P <0.01 ). It was only 37. 0% , 29. 8% , 30. 0% of that in control group, respectively. Conclusion Phenylalanine might interfere the growth of axons, dendrites, synapses in the cerebral cortex through influencing the expression of Rho GTPases mRNA, which probably is the mechanism of mental retardation associated with phenylke-tonuria.
