葡萄籽提取物对小鼠急性化学性肝损伤保护作用的实验研究

摘    要:
目的:研究葡萄籽提取物(GSE)对四氯化碳(CCl4)所致小鼠急性化学性肝损伤的保护作用,并探讨其可能的作用机制,为进一步研究GSE的保健功能奠定基础。方法:雄性昆明种小鼠60只,随机分为正常对照组、急性化学性肝损伤模型组、联苯双酯阳性对照组(150mg/kg)以及GSE低、中、高剂量组(50、250、500mg/kg)共6组。测定并比较各组小鼠肝脏和脾脏系数,采用赖氏法测定血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性,采用黄嘌呤氧化酶法测定肝组织中超氧化物歧化酶(SOD)活性,采用硫代巴比妥酸法(TBA)测定肝组织中丙二醛(MDA)含量,并对各组进行比较,观察肝脏病理组织学变化。结果:GSE各剂量组均能升高急性化学性肝损伤小鼠血清ALT和AST活性(P〈O.01),升高肝组织SOD活性(P〈O.01),降低MDA含量(P〈O.01),并能不同程度地改善肝脏病理组织损伤。结论:葡萄籽提取物对CCl4所致急性化学性肝损伤具有保护作用。

Abstract:
Objective:To study the protective effects of grape seed extracts(GSE) against the acute chemical liver injury in mice and research its possible mechanism for offering scientific basis of its health function.Methods:Randomly distribute 60 male Kunming mice to control group,acute chemical liver injury model group,DDB group(150 mg/kg) and low,middle,high dose groups of GSE(50,250,500 mg/kg).Determination and comparing of liver and spleen index between groups,using Reitman-Frankel to assay serum ALT and AST levels,xanthine oxidase to assay liver tissue SOD activities and thiobarburic acid to assay MDA content in liver tissue,and observing histopathological change of liver.Results:Treatment with different dose GSE could significantly increase serum ALT,AST levels(P<0.01) and liver tissue SOD activities(P<0.01),decrease MDA content in liver tissue(P<0.01).Histopathological observations showed that GSE could ameliorate the liver injury in different degree.Conclusions:GSE showed protective effects on the acute chemical liver injury induce by carbon tetrachloride in mice.
