摘 要: | 目的观察鹿茸多肽对人胶质瘤细胞生长抑制率及细胞周期的影响,探讨鹿茸多肽抑制肿瘤细胞的增殖及调节细胞周期作用的机制。方法在体外传代的培养人胶质瘤细胞(U251)细胞株中加入不同浓度(50、100、200、400、800μg/mL)的鹿茸多肽,诱导培养48 h,镜下观察细胞的形态学变化,MTT法检测鹿茸多肽对肿瘤细胞增殖情况,计算抑制率;流式细胞术(FCM)分析鹿茸多肽,诱导培养肿瘤细胞48 h后,细胞周期变化情况。结果形态学观察表明,与对照组相比,随着药物剂量的增加,实验组细胞密度逐渐降低,细胞间隙逐渐增宽,细胞体积逐渐缩小,细胞内颗粒逐渐增多。MTT法检测结果表明,鹿茸多肽对肿瘤细胞的增殖抑制作用明显,细胞生长抑制率随着剂量增加抑制率逐渐增加,呈现出良好的剂量依赖性。FCM检测表明,肿瘤细胞G0/G1期与S期所占百分比逐渐减少,G2/M期阻滞。48 h均出现凋亡峰,及凋亡细胞群。结论鹿茸多肽对体外培养的人胶质瘤细胞的增殖有抑制作用,并具有量效关系。可使肿瘤细胞阻滞在细胞周期的G2/M期,继而引起的细胞周期阻滞和诱导其凋亡。

Abstract: | Objective To observe the effects of velvet antler polypeptides on the inhibition rate of growth of human glioma cell and cell cycle and to explore the mechanism of inhibition of velvet antler polypeptides on the proliferation of tumor cells and the regulation of cell cycle.Methods Glioma cells(U251) of human cultured in vitro were added into different concentrations of velvet antler extract(50,100,200,400,800 μg /mL) and were induced cultured for 48 h,and then the changes of cell morphology were observed by microscope.MTT method was used to determine the effects of velvet antler polypeptide on tumor cell proliferation condition and inhibition rate was calculated.Flow cytometry(FCM) was applied to analyze changes of cell cycle after 48 h of induced culture of tumor cells by velvet antler polypeptides.Results Morphology observation results show that compared with the control group,cell density decreased gradually,intercellular space gradually broadened,cell volume shrank gradually,intracellular particles increased gradually in the experimental group with the increase of drug dose.MTT detection results show that the inhibition function of velvet antler polypeptides on tumor cell proliferation,inhibition rate of cell growth increased gradually with the increase of dose,presenting a favorable dose-dependency.FCM test showed that the percentage of G0 /G1 phase and S phase in tumor cells gradually decreased,and G2 /M phase was retarded.Apoptosis peak and apoptosis of cells appeared after 48 h.Conclusion Velvet antler polypeptides can inhibit the proliferation of human glioma cell of human cultured in vitro and had dose-effect relationship.It can make the tumor cell block in G2 /M phase in the cell cycle,which in turn caused cell cycle arrest and induce its apoptosis. |
