兔动脉粥样硬化斑块处单核细胞趋化蛋白-1的表达

摘    要:目的了解单核细胞趋化蛋白-1(monocyte chemoattractant protein-1,MCP-1)在动脉粥样硬化中的作用。方法采用免疫组化、双抗夹心ELISA和原位杂交的方法,检测MCP-1蛋白在免动脉粥样硬化斑块处的表达及定位MCP-1mRNA的表达。结果8、12周模型组内膜显著增厚并有明显阳性染色,半定量分析动脉粥样硬化模型组阳性染色的面积分别是对照组的2.04、3.32倍,平均积分光密度(IOD)值分别是对照组的2.33、3.43倍。8、12周动脉粥样硬化模型组MCP-1蛋白含量与总蛋白比值和对照组比较均有显著性差异,分别是对照组的2.90、3.58倍。8周模型组血管内膜有强的阳性染色,MCP-1mRNA的表达增加。结论MCP-1的蛋白和基因上调主要是在高脂血症兔动脉粥样硬化模型的脂纹期。MC-1参与动脉粥样硬化斑块的形成,主要在动脉粥样硬化的早期。

Abstract:Objective To establish the animal model of hyperlipemia in rabbits and investigate the changes of protein and gene expression of monocyte chemoattractant protein-1(MCP-1) in the plaques of atherosclerosis and explore the temporal correlation between MCP-1 expression and plaque development.Methods A total of 30 New Zealand rabbits were randomly assigned into feed with ordinary diet and high lipid diet.In the rabbits with high lipid diet,250 mg/kg bovine serum albumin was injected before animal model establishment,then every 15 d once more.The MCP-1 expression in the atherosclerosis plaques in aorta arch was detected by immunohistochemistry,ELISA,in situ hybridization on week 4,8,12 after hyperlipemia rabbits were sacrificed.Results Immunohistochemical results showed the expression of MCP-1 protein increased obviously in intima of hyperlipemia rabbits on week 8 and 12.Quantitative analysis showed MCP-1 positive area of atherosclerosis model was 2.04 times and 3.32 times as large as that of control group on week 8 and 12.The value of IOD of atherosclerosis model was 2.33 times and 3.43 times as high as that of control group on week 8 and 12.ELISA results showed the ratio of the MCP-1 protein to total protein of atherosclerosis model was 2.90 times and 3.58 times as high as that of control group on week 8 and 12.In situ hybridization showed positive location was strong in intima of hyperlipemia rabbits on week 8.Conclusion MCP-1 protein and gene expression was up-regulated in the development of fatty streaks in hyperlipemia rabbits.MCP-1 plays an important role in the early atherosclerotic lesions.
