摘 要: | 目的:探讨雷公藤红素对人肝癌MHCC97H细胞侵袭转移的抑制作用及相关机制。方法:应用Transwell技术观察不同浓度(2.5μmol·L^-1、5.0μmol·L^-1、10.0μmol·L^-1)雷公藤红素对MHCC97H细胞侵袭力和迁移力的影响。Westernblot法检测雷公藤红素处理后对C-Jun氨基末端激酶(JNK)信号通路及NF-κB/p65核浆分布变化的影响。结果:雷公藤红素处理后24 h,迁移实验及侵袭实验结果显示,加药组透膜细胞数较对照组明显减少(P〈0.05)。Westerblot结果显示磷酸化JNK表达上调,NF-κB/p65入核减少。结论:雷公藤红素体外具有抑制人肝癌细胞侵袭和转移的作用,其机制可能与活化JNK通路、抑制NF-κBp65入核有关。

Abstract: | Objective:To explore the efect of celastrol on the invasion and metastasis of human liver cancer MHCC97H cells and related mechanism. Methods:The transwell chamber was used to investigate the invasion and migration ability of MHCC97H cells after treated with celastrol (2.5,5 and 10 μmol · L^-1 ) for 24 hours. The expression and phosphorylation level of JNK signaling pathway, and subcellular distributions of NF-κB/p65 were all detected through Western blot. Results:After treated with different concentrations of celastrol for 24 h,The membrane-permeating MHCC97H cells in invasion and migration assay were decreased in celastrol treated group comparied with control group. Western blot showed that the expression of p-JNK was increased but total JNK protein were basically unchanged, and expression of NF-κB/p65 decreased in nuclei and increased in the cytoplasm. Conclusion:Celastrol inhibited cell invasion and migration in MHCC97H cells in vitro. The possible mechanism may be related to up-regulation of p-JNK expression, and expression of NF-κB/p65 decreased in nuclei. |
