柴芩肾安方对IgA肾病大鼠肾小球足细胞podocin表达的影响

目的观察柴芩肾安方对IgA肾病（IgAN）大鼠肾小球足细胞podoc in表达的影响。方法雄性SD大鼠随机分为正常组、切肾组、IgAN组、柴芩肾安方组和氯沙坦组,通过切除单侧肾并反复静脉注射葡萄球菌肠毒素B（SEB）建立大鼠IgAN模型,分别进行药物干预。第12周末,检测各组大鼠24h尿蛋白量（Upr）,观察肾小球病理变化,并采用免疫组化和实时定量PCR检测足细胞podo-c in蛋白及mRNA的表达。结果与正常组比较,IgAN组大鼠Upr、肾小球系膜细胞、系膜基质相对含量均显著升高（P〈0.01）,大部分足突融合,足细胞podoc in蛋白及mRNA表达均明显下调（P〈0.01）。柴芩肾安方组上述指标均明显改善,与IgAN组比较差异显著（P〈0.01）,且其podoc in蛋白表达高于氯沙坦组（P〈0.01）。结论柴芩肾安方能减轻IgAN大鼠蛋白尿及肾小球病变,这可能与其恢复足细胞podoc in表达有关。

Effects of "Chaiqin Shenan Decoction" on expression of podocin in glomerular podocytes in IgA nephropathy rats

Objective To observe the effects of ＂Chaiqin Shenan Decoction＂ on expression of podocin in glomerular podocytes in IgA nephropathy（IgAN） rats.Methods The male SD rats were randomly divided into five groups： normal group,model group,IgAN group,herb group and Losartan group.IgAN was established by unilateral nephrectomy and repeatedly injecting steophylocdccus enterotoxin B（SEB）.All groups were treated accordingly.At the end of 12th week,the levels of 24-hour urinary protein（Upr） were measured;the pathological changes of glomeruli were observed,and the immunohistochemistry and real-time quantitative PCR were used respectively to examine the protein and mRNA expression of podocin in glomeruli.Results Compared with the normal group,Upr,mesangial cells and mesangial matrix were increased significantly in IgAN group（P0.01）;foot processes fusion was observed,and the expression of protein podocin and expression of mRNA in glomerular podocytes were reduced significantly（P0.01）.Compared with the IgAN group,all the indicators above improved remarkably in herb group（P0.01）;compared with the losartan group,the protein expression of podocin was higher in the herb group（P0.01）.Conclusion ＂Chaiqin Shenan Decoction＂ can relieve the proteinuria and glomerulopathy in rats of IgA nephropathy by elevating the podocin expression of podocyte.
