Abstract:Objective To study the influence of 3′-azido-3′-deoxythymidine(AZT) on the apoptosis of gastric cancer cell line SCG 7901 and the expressions of BAX protein and proliferation cell nuclear antigen(PCNA) and explore the mechanisms.Methods The gastric cancer cell line SCG 7901 treated with 1.0 mmol·L-1 AZT for 6 d were used as AZT group,at the same time,control group was set up.The apoptotic rates of SGC 7901 in two groups were detected with MG-P-MY staining.The expressions of Bax protein and PCNA of gastric cancer cell line SCG 7901 6 d after drug administration were determined by immunohistochemical technique(EnVision).Results The apoptotic rates in AZT proup and control group were(16.421±0.713)% and(3.366±0.271)% respectively,the apoptotic rate in AZT group was significantly higher than that in control group(P<0.05).Compared with control group,the expression of Bax protein was significantly increased(16.851%±10.064% vs 47.389%±18.042%,P<0.05) and the expression of PCNA was significantly decreased(23.446%±13.055% vs 62.332±22.767%,P<0.05) in AZT group.Conclusion AZT can induce the apoptosis of gastric cancer cell line SCG 7901,promote the expression of Bax protein and inhibit the expression of PCNA.
