白藜芦醇对帕金森病模型大鼠氧化应激损伤的拮抗效应

摘    要:目的探讨白藜芦醇对帕金森病(Parkinson’s disease,PD)模型大鼠氧化应激损伤的拮抗效应。方法应用6-OHDA纹状体内注射制作PD模型大鼠,将模型成功大鼠随机分为4组:模型组,白藜芦醇10,20,40mg/kg/d治疗组以及ViLE对照组(50mg/kg/d),并设假手术组及正常给药组(自藜芦醇20mg/kg/d),每日口服给药一次,连续10周,给药过程中每周进行一次行为学检测,然后利用化学比色法检测损毁侧脑组织中SOD、GSH—PX活性以及MDA含量。结果1.白藜芦醇治疗组可使大鼠旋转行为明显改善;2.白藜芦醇治疗组可明显增加SOD、GSH—PX活性,降低MDA含量。结论白藜芦醇对帕金森病模型大鼠具有保护作用,其机制与白藜芦醇减轻6-OHDA所致的氧化应激损伤有关。

Abstract:Objective To explore antagonistic effects of resveratrol on Parkinson' s disease (PD) model rats. Methods The PD model rats were indueed by injecting 6-OHDA into the right striatum, The valid PD rats were randomly divided into four groups: model control group, resveratrol groups (10, 20 and 40 mg/kg/d), the VitE control group (50 mg/kg,/d), sham group and normal administration group (resveratrol 20 mg/kg/d). Resveratrol was given orally once daily for 10 weeks, Rotational test was examined weekly during administration, then the activity of SOD, GSH-PX and level of MDA in injured brain tissues were detected respectively with chemical chromatometry. Results 1. Resveratrol can ameliorate significantly turns of rats. 2. Resveratrol can increase significantly the average activity of SOD, GSH-PX, and decrease obviousely the average level of MDA. Conclusion Resveratrot have obvious protective effect on PD model rats, which may be related to attenuate 6-OHDA-indueed oxidative stress injury.
