预刺激小脑顶核对脑缺血神经元凋亡的防治作用

摘    要:目的探讨预刺激脑缺血大鼠小脑顶核对神经元凋亡的防治作用。方法采用线栓法大鼠大脑中动脉栓塞模型,缺血时间均为1.5h/再灌注24h;于缺血前1、4、7d分别刺激小脑顶核、齿状核1h。以尾状核冠状切面作为观察对象,应用TUNEL法观察对照组、假手术组、刺激小脑顶核和齿状核组凋亡神经元的表达情况。结果缺血前1、4、7d刺激小脑齿状核各组、单纯缺血/再灌注组、假手术组TUNEL阳性细胞数比较无显著性差异(P>0.05),而缺血前1、4、7d预刺激小脑顶核能明显抑制TUNEL阳性细胞数(P<0.05)。结论预先电刺激小脑顶核的缺血脑保护作用可能与其抑制神经元凋亡有关。

Abstract:Objective To investigate whether electrical stimulation to the cerebella fastigial nucleus(FN) for 1h can effect apoptosis before MCAO/R.Method A model of the middle cerebral artery occlusion and reperfusion(MCAO/R) in Wistar rats was performed with the intraluminal filament occlusion.The rats brains were cut in the coronal planes at the caudoputamen as the templates.the FN and DN(Dentate Nucleus,DN) were stimulated for 1h either 1d before MCAO/R (1.5 h/24 h) or 4 d,or 7 d.The TUNEL staining was used to facilitate the observation of the distributions and quantities of apoptosis in the brain tissues following the MCAO/R groups.Results There were no significant discrepancy of the apoptotic cells among the smple ischemia/reperfusion group,the sham operation group(FN was no stimulated) ischemia/reperfusion group and the DN stimulationgroups(P >0.05), they were signigicantly inhibited by the FN stimulation(P< 0.05).Conclusions The protective effect of FN pre stimulation on cerebral ischemic probably relates to the inhibition of the quantities of apoptotic cells.
