硫酸软骨素对不同时期液氮冷冻保存的家兔心脏瓣膜的影响

摘    要:目的观察含硫酸软骨素(chondroitin sulfate,CS)的液氮冷冻保存的同种异体心脏瓣膜在不同保存时间内细胞活性及瓣膜组织学结构的变化。方法取48枚兔主动脉瓣随机分成6组,每组8只并经灭菌处理。对照组于灭菌后立即行瓣膜细胞活性测定及组织学检查;实验Ⅰ~Ⅴ组分别在含有硫酸软骨素的保护液中液氮深低温保存1、3、5、7、9个月后取出,复温后行瓣膜细胞活性测定、光镜及电镜检查。结果实验Ⅰ~Ⅴ组D(490)值分别为(0.073±0.014)、(0.067±0.021)、(0.066±0.015)、(0.063±0.020)、(0.042±0.018),与对照组(0.105±0.029)比较,瓣膜细胞活性明显下降(P<0.05);实验Ⅰ~Ⅳ组间,各组瓣膜细胞活性无明显差异(P>0.05);实验Ⅴ组与实验Ⅰ组比较,瓣膜细胞活性明显下降(P<0.05)。各实验组瓣膜组织学结构保存尚好。结论CS能维持液氮深低温中保存的心脏瓣膜的细胞活性,使冷冻保存7个月内细胞活性无明显下降,组织学结构保存较好,瓣膜可用性大。
