6,7-二甲氧基香豆素抗急性肾功能衰竭的作用机制研究

摘    要:目的 探讨6 ,7 二甲氧基香豆素抗急性肾功能衰竭( A R F) 的作用,并初步分析其作用机制。方法 观察6 ,7 二甲氧基香豆素对内毒素诱导小鼠 A R F 模型的影响,分析其对 A R F 的作用;观察消炎痛或 L N A M E 协同 N E 升高离体灌流肾肾灌注压以分析6 ,7 二甲氧基香豆素对 P G I2 和 N O的影响。结果 6 ,7 二甲氧基香豆素对内毒素引起的小鼠急性肾衰有明显的拮抗作用,能降低血清肌酐浓度、血清尿素氮浓度、肾匀浆 S O D 活性及肾匀浆 L P O 含量的升高,减轻肾小球毛细血管及肾间质充血。6 ,7 二甲氧基香豆素有延缓正常灌流肾肾灌注压缓慢上升的作用。呈剂量依赖性拮抗 N E 引起的肾灌注压的升高。通过 P G I2 及 N O 的释放等减小肾血管阻力。结论 6 ,7 二甲氧基香豆素有抗内毒素诱导的 A R F 的作用,主要通过防止氧自由基的生成及( 或) 清除已生成的氧自由基,促使血管内皮细胞释放 N O 和 P G I2 ,减少肾血管阻力等综合作用实现

Abstract:AIM To investigate the effect and mechanism of 6,7 dimethoxycoumarin on the acute renal failure (ARF). METHODS In the model of endotoxin induced ARF in mice, the variation of the concentration of Cr and BUN in serum and SOD, LPO and GSH in renal homogenate was detected, and the role of 6,7 dimethoxycoumarin on the PGI 2 and NO was studied through the isolated perfused kidney. RESULTS 6,7 dimethoxycoumarin not only decreased the abnormal higher concentration of Cr and BUN but also improved activity of SOD, reduced the quantity of LPO without influence on the activity of GSH and ameliorated the congestion of the kidney in endotoxin induced ARF mice, which indicated its anti ARF effect through scavenging reactive oxygen species and correcting their injury. 6,7 dimethoxycoumarin delayed the increase of the normal pressure and inhibited the NE induced increased pressure in concentration dependent manner. 6,7 dimethoxycoumarin could also enhance the production of PGI2 and NO. CONCLUSION 6,7 dimethoxycoumarin contributed to the cure of acute renal failure and was one of the active substances in Kangjue Injection.
