山莨菪碱对沙土鼠脑缺血后延迟性神经元死亡的影响

摘    要:目的 研究山莨菪碱对沙土鼠脑缺血后海马延迟性神经元死亡的影响并探讨其与纹状体多巴胺和 A T P 含量变化的关系。方法 制备沙土鼠前脑缺血再灌注模型,脑缺血10 min 。32 只沙土鼠随机分为假手术组、脑缺血组、脑缺血再灌注组和山莨菪碱组。应用病理检查判断脑缺血后延迟性神经元死亡情况,高效液相测定纹状体 A T P 和多巴胺的含量。结果 脑缺血组沙土鼠纹状体 A T P 和多巴胺含量明显低于假手术组。在再灌注60 min 时,山莨菪碱组沙土鼠纹状体多巴胺和 A T P 含量明显高于脑缺血再灌注组[ 多巴胺:(59 ±10) vs (35 ±14) mg·kg - 1 , P< 001 ; A T P:(082 ±012) vs (062 ±010) m mol·kg- 1 , P< 005] 。山莨菪碱组沙土鼠脑缺血后海马 C A1 区延迟性神经元死亡数目明显少于脑缺血再灌注组。结论 山莨菪碱可明显减少沙土鼠脑缺血后延迟性神经元死亡数目,机制可能与其减少脑缺血再灌注期间纹状体多巴胺释放和促进 A T P 含量恢复的作用有关

Abstract:AIM To study the effect of anisodamine on delayed neuronal death (DND) and to investigate the relationship between DND and the changes of ATP, DA(dopamine) contents in striatum during forebrain Ischemia/reperfusion in gerbils. METHODS To prepare the model of forebrain ischemia/reperfuison in gerbils and the insult time was 10 min. 32 gerbils were divided into 4 groups, Sham operated group, cerebral ischemic group, cerebral ischemia/reperfusion group and anisodamine group. The DND was assessed by histological examination, and the content of ATP and DA in striatum was determined by high performance liquid chromography(HPLC). RESULTS The contents of ATP and DA in striatum in cerebral ischemic group was much lower than those in sham operated group. However, the content of ATP and DA in striatum in anisodamien group by the end of 60 min was significantly higher than those in cerebral ischemia/reperfusion group, [DA:(5 9±1 0) vs (3 5±1 4)mg·kg -1 , P <0 01; ATP:(0 82±0 12) vs (0 60±0 10)mmol·kg -1 , P <0 05]. Anisodamine could also remarkably reduce the DND in hippocampal CA 1 sector. CONCLUSION Anisodemine can decreased DND after cerebral ischemia in gerbils, and the mechanism may be related to its role of inhibiting DA release and quickening the restoration of ATP content in striatum.
