摘    要: 目的观察羟乙葛根素对大鼠脑缺血再灌注损伤后脑组织ICAM-1及VCAM-1表达的影响,以探讨其作用机制。方法线栓法制备大鼠局灶性脑缺血再灌注损伤模型,大鼠脑缺血2 h再灌注24 h后,采用Western blot及RT-PCR法观察大鼠脑组织ICAM-1、VCAM-1的蛋白及mRNA表达情况。结果羟乙葛根素可明显降低缺血区脑组织ICAM-1、VCAM-1的蛋白及mRNA表达。结论羟乙葛根素可能通过降低缺血区脑组织黏附分子表达发挥其脑保护作用。

Abstract:OBJECTIVE To investigate the effects of hydroxyethylpuerarin on expression of ICAM-1 and VCAM-1 in rats with focal cerebral ischemia-reperfusion injury and explore its mechanism of action.METHODS Rats were subjected to cerebral ischemia-reperfusion injury by middle cerebral artery occlusion(MCAO).After 2 h ischemia and 24 h reperfusion,the protein and mRNA expression of ICAM-1 and VCAM-1 were evaluated by Western blot and RT-PCR analysis.RESULTS Hydroxyethylpuerarin significantly reduced the expression of ICAM-1 and VCAM-1 in ischemic brain tissue after cerebral ischemia-reperfusion.CONCLUSION Hydroxyethylpuerarin may attenuate ischemic brain injury by down-regulation the expression of adhesion molecules in ischemic brain tissue.
