摘    要:目的:采用HPLC法测定射干根茎与须根中鸢尾苷的含量,并比较两者含量。方法采用 Eclipse XDB-C18色谱柱(250mm×4.6mm,5μm),以乙腈-0.2%磷酸溶液(20∶80)为流动相,流速1.0mL·min -1,检测波长266nm,进样量20μL,柱温35℃。结果鸢尾苷进样量在3.1875~47μg范围内与峰面积线性关系良好,r=0.9998,精密度、重复性、稳定性良好,平均加样回收率为99.81%。结论所建方法可同时测定射干根茎与须根中鸢尾苷的含量,并且根茎中的鸢尾苷含量显著高于须根。

Abstract:OBJECTIVE To establish a HPLC method for determining the content of tectoridin in Belamcanda Chinensis and to compare the content of tectoridin in rhizome and beard.METHODS The assay was performed on a Eclipse XDB-C18 column (250mm ×4.6mm,5μm);eluted with a mobile phase of acetonitrile-0.2%phosphoric acid (20∶80) at a flow rate of 1.0mL·min-1;The detection wavelength was 266nm and the column temperature was 35℃.Results The linear range of irisflorentin was 3.1875 -47μg ( r =0.9998 ).The reproducibility and stability were good and the average recovery was 99.81%.CONCLUSION The method is simple、rapid and reliable.The content of tectoridin in rhizome is by far higher than that in beard.
