摘    要:目的探讨2型糖尿病大鼠肾脏病变与局部能量代谢的关系。方法4只Wistar大鼠为正常对照组,10只GK Wistar大鼠随机分为糖尿病对照组和糖尿病硫辛酸治疗组(腹腔内隔日注射α-硫辛酸35 mg.kg-1),各5只。12周后测定24 h尿蛋白排泄率、肾组织ATP酶活性和氧化应激指标。结果糖尿病对照组大鼠尿蛋白排泄率增高,肾组织ATP酶活性降低,以及MDA水平升高和GSH水平降低,而硫辛酸治疗能明显改善上述异常变化。24 h尿蛋白排泄率与SOD、GSH和ATP酶活性呈显著负相关,肾组织ATP酶活性与GSH呈显著正相关。结论2型糖尿病大鼠肾脏存在明显的线粒体功能障碍,此与肾病的发生和发展有关。

Abstract:Objective To evaluate the relationship of renal lesion to mitochondrial function of the kidney in type 2 diabetic Goto Kakisaki rats. Methods Four healthy Wistar rats were assigned as normal control group, and 10 GK rats were randomly divided into two groups:diabetic control group and lipoic acid-treated diabetic group ( α-lipoic acid 35 mg/kg intraperitoneal injection qd alt), with 5 GK rats in each group. The treatment program was for a period of 12 weeks, during which urine protein excretion and ATPase activity in renal homogenate were measured. Results Compared with normal control group, there were significant decrease in the ATPase activity and GSH level, and increase in urine protein excretion and MDA level in diabetic control group, and these changes were alleviated in the α-lipoic acid treated group. There were negative correlations between urine protein excretion and SOD level, GSH level and ATPase activity, and a positive correlation between ATPase activity and GSH level in renal tissues. Conclusion In type 2 diabetic rats, there is renal mitochondrial dysfunction which contributes to the development of diabetic nephropathy.
