摘    要:
目的 探讨原发性肝细胞癌(HCC)中诱骗受体3(DcR3)表达和意义及与细胞凋亡的关系.方法 应用免疫组织化学Envision二步法和TUNEL技术对62例HCC组织和配对癌旁组织、15例正常肝组织进行DcR3和细胞凋亡检测.结果 44例HCC中DcR3表达、阳性表达率为70.97%,配对癌旁组织为20.96%,正常肝组织无阳性表达,HCC中DcR3表达明显高于其他组(P<0.01),门静脉癌栓形成组中DcR3表达高于阴性组(P<0.01).HCC、癌旁和正常肝组织凋亡指数(AI)分别为3.52±1.65、6.08±1.71和6.87±1.78,HCC与后两者比较差异有统计学意义(P<0.01).DcR3阳性表达组中AI明显低于阴性组(P<0.01).结论 DcR3在HCC呈稳定高表达,与癌细胞凋亡相关,可能参与HCC的发生和影响肿瘤的预后.

Expression of decoy receptor 3 in hepatocellular carcinoma and its correlation to apoptosis
Abstract:
Objective To evaluate the expression of decoy receptor 3 (DcR 3) in hepatocellular carcinoma and its correlation to apoptosis. Methods Immunohistochemical thechniques (two-step Envi-sion) were used to detect the expression of DcR 3 in 15 normal liver specimens ,62 heptocellular carcino-ma specimens and corresponding paracancerous specimens. Terminal deoxynucleotidyl transferase-mediated dUTP digoxigenin nick and labeling (TUNEL) was used to identify the apoptosis in tissues. Results Ex-pression of DcR 3 in HCC (70.97% ,44/62) was significantly higher than that in corresponding paracan-cerous specimens (20.96%, 13/62 ,P < 0. 01 ) and that in normal liver specimens (0,0/15, P < 0.01 ) , and HCC with tumor thrombus of portal vein showed stronger DcR 3 expression. Apoptosis index (AI) in HCC specimens ( 3.52 ± 1.65 ) was significantly higher than that in corresponding paracancerous speci-mens (6.08 ± 1.71,P < 0. 01 ) and that in normal liver specimens (6.87 ± 1.78, P < 0. 01 ). As com-pared to HCC without DeR 3 expression, AI was significantly lower in HCC positive for DcR 3 (P < 0. 01 ). Conclusion Our results demonstrate that DcR 3 is overexpressed in HCC and associated with ap-optosis. DcR 3 may play an important role in the progression of HCC and examination of its expression might be helpful for diagnosis and treatment of HCC.
