制何首乌对肝脏生化物质影响的实验研究

摘    要:
目的实验研究不同剂量的陕西安康野生制何首乌对大鼠肝脏的损伤。方法每天按不同剂量,分别相当于成人每日12g临床用量的20、50、100、200倍灌服大鼠1次。连续给药90d,恢复期15d。分别于30d、60d、90d及105d采取动物血液进行生化检测,同时在90d时摘取大鼠肝脏匀浆进行丙二醛(MDA)、单胺氧化酶(MAO)、谷胱甘肽-S转移酶(GSH-ST)检测。结果给药组与对照组相比,大剂量(40g/Kg/d)和较大剂量(22g/Kg/d)给药组对大鼠肝脏转氨酶AST在90d有一定的影响(P〈0.05),MDA有显著性升高(P〈0.05)。结论制何首乌长期灌胃大剂量(40g/kg/d)对大鼠肝脏有轻度的肝损害,常用剂量(22g/kg/d)各阶段应用则未见毒副作用。

Abstract:
Objective: To study different doses of wild Radix polygonl multiflori preparata's(vegetated in Ankang,Shaanxi) injuring effects on rat's liver Methods: The rats were drenched with different dosages each day,that is 20 times,50 times,100 times,200 times of the amount of an adult's clinical dosage-12 g respectively.It was successively administered for 90 days,and the recovery period was 15 days.The rats' blood samples were taken on the 30 d,60 d,90 d,and 105 d respectively for biochemical examination.Meanwhile,the rats' liver homogenate was taken on the 90 d to do MDA,MAO and GSH-ST detection.Results: Compared with the control group,the experimental groups which were given 40g/Kg/d and 22g/Kg/d,the medicine had some effects on rat liver's transaminase AST in 90d(P<0.05),MDA increased significantly(P<0.05).Conclusion: Long term use of high dose Radix polygonl multiflori preparata(40g/kg/d)for intragastric administration does minor injure to rat's liver.No toxic and side effects were seen on each stage when used at normal dosage(22g/kg/d).
