雪旺细胞与神经元共培养时NGF、CNTF和GDNF mRNA共表达的计算机成像

摘    要:目的研究雪旺细胞与背根节神经元共培养条件下多种神经营养因子共表达的计算机数字成像技术.方法运用多色荧光原位杂交技术和计算机图像处理技术, 对相同视野下分别采用绿、红、蓝单色滤光片摄取的NGF(绿)、CNTF(红)和GDNF(蓝)mRNA表达的荧光图像以及明视场细胞图像进行其共表达分布的图像合成.结果在单个雪旺细胞内同时显示出NGF、CNTF和GDNF mRNA表达所对应绿、红、蓝色区域及其在细胞相同部位同时表达所对应的由绿、红、蓝色混合而衍生的白、黄、紫红和浅蓝色区域.结论应用多色荧光原位杂交技术和计算机图像处理技术可对雪旺细胞中多种神经营养因子的共表达进行可视化研究.

Abstract:Objective To study the computerized digital imaging of the co-expression of multiple neurotrophic factors in Schwann cells cocultured with neurons from dorsal ganglion. Methods The green, red and blue fluorescent images respectively corresponding to the expression of NGF, CNTF and GDNF mRNA and the image of Schwann cells under bright field were composed into one image which can reveal the co-expression of above 3 factors simultaneously by means of multicolor fluorescent in situ hybridization and computer image processing technique. Results The green, red and blue area corresponding to the expression of NGF, CNTF and GDNF mRNA and the white, yellow, mauve and azury area corresponding to the co-expression of any 2 or 3 factors in the same site resulting from the various combination of red, green and blue color were showed on the level of single schwann cell. Conclusion The visualized study of the co-expressions of multiple neurotrophic factors in Schwann cells is available.
