Abstract:
Objective It is to investigate the intervention effect of Huangbai capsule combined with olotatidine hydrochloride on acute eczema model rats and its anti-allergic mechanism.Methods Fifty SD rats were randomly divided into control group,model group,Huangbai capsule group,olotatidine hydrochloride group and combination group,each group had 10 rats.Except for the control group,the acute eczema rats models were established by applying 2,4-dinitrochlorobenzene(DNCB)on the right back.On the 10th day after model establishment,the control group and model group were given 0.9%saline by gavage,the Huangbai capsule group was given Huangbai capsule by gavage,the olotatidine hydrochloride group was given olotatidine hydrochloride by gavage,and the combination group was given Huangbai capsule and olotatidine hydrochloride by gavage,all one time per day,continuously treated for 10 days.The score of eczema,skin swelling were evaluated and the He staining histopathological changes were observed on the 20th day after model establishment.The levels of serum inflammatory factor interferon-gamma(IFN-γ),interleukin-4(IL-4)and tumor necrosis factor-alpha(TNF-α)were detected by ELISA.The protein and mRNA expression levels of intercellular adhesion molecule-1(ICAM-1)and vascular endothelial cell adhesion molecule-1(VCAM-1)were detected by Western blot and RT-PCR.Results The score of eczema and skin swelling and the levels of serum IFN-γ,IL-4 and TNF-αin Huangbai capsule group,olotatidine hydrochloride group and combination group were significantly lower and the pathological morphology were significantly improved compared with model group,the scores and levels in the combination group were lower than those in Huangbai capsule group and olotatidine hydrochloride group(P<0.05).The protein and mRNA expression levels of ICAM-1 and VCAM-1 in the Huangbai capsule group,olotatidine hydrochloride group and combination group were significantly lower than those in the model group(P<0.05),and the combination group was significantly lower than the Huangbai capsule group and olotatidine hydrochloride group.Conclusion Huangbai capsule combined with olotatidine hydrochloride can reduce the eczema score and skin swelling,improve the histopathology of acute eczema rats,which may be related to the reduction of inflammatory reaction and the reduction of ICAM-1 and VCAM-1 protein and mRNA expression.
