摘 要: | 目的采用生物信息分析技术探讨胶质母细胞瘤中和长链非编码RNA GAS5相关的m6A基因的筛选及临床特征分析。方法从开放的癌症基因图谱(TCGA)和基因型组织表达(GTEx)数据平台下载脑胶质瘤样本转录水平数据。根据TCGA胶质母细胞瘤患者GAS5基因的表达量做相关性分析,筛选出GAS5相关m6A基因。通过权重基因共表达网络分析(WGCNA)构建加权基因共表达网络,寻找协同表达的基因模块,筛选GAS5共表达的基因。GAS5共表达的基因和GAS5相关m6A基因取交集。CGGA数据库分析筛选出的基因与胶质瘤患者的临床特征及预后之间的关系。结果分析的13个m6A基因中,相关性系数> 0.4,P<0.001,7个基因(HNRNPC、FTO、KIAA1429、ALKBH5、YTHDF2、WTAP、YTHDC2)与GAS5相关,其中HNRNPC、KIAA1429、YTHDF2、WTAP呈正相关,FTO、ALKBH5、YTHDC2呈负相关。通过构建WGCNA网络,筛选GAS5共表达的基因,brown模块共1 292个基因与GAS5相关性最高(cor=0.84,P=1e-200)。HNRNP...

Abstract: | Objective To explore the M6A gene associated with long non-coding RNA GAS5 in glioblastoma by using bioinformatics analysis technology and to analyze its clinical characteristics.Methods Transcriptional data of glioma samples were downloaded from the open Cancer Genome Atlas(TCGA)and Genotype Tissue Expression(GTEX)data platforms.According to the expression level of GAS5 gene in TCGA glioblastoma patients,correlation analysis was conducted to screen out GAS5-related M6A gene.Weighted gene co-expression network analysis(WGCNA)analysis was used to construct a weighted gene co-expression network to search for coexpressed gene modules and screen genes co-expressed in GAS5.The GAS5 co-expressed gene intersects with GAS5 related M6a gene.CGGA database was used to analyze the relationship between the screened genes and the clinical characteristics and prognosis of glioma patients.Results Among the 13 m6A genes analyzed,correlation coefficient>0.4,P<0.001.7 genes(HNRNP,FTO,KIAA1429,ALKBH5,YTHDF2,WTAP,YTHDC2)were related to GAS5.HNRNPC,KIAA1429,YTHDF2 and WTAP were positively correlated,while FTO,ALKBH5 and YTHDC2 were negatively correlated.Through the construction of WGCNA network,genes co-expressed by GAS5 were screened,and a total of 1292 genes in Brown module had the highest correlation with GAS5(cor=0.84,P=1e-200).HNRNPC was an intersection gene co-expressed by GAS5,M6A related genes and GAS5.The differences in HNRNPC gene expression were statistically significant in IDH Mutation(P=0.008),IDH Mutation status&1p/19q Codel status(P=2.1e-05).In the case of high expression of GAS5,high expression of HNRNPC was conducive to survival(P<0.05).Conclusions M6A related gene HNRNPC is closely related to GAS5 in glioblastoma.HNRNPC was associated with malignant degree and prognosis of glioblastoma.Further elucidation of the role and mechanism of HNRNPC in the occurrence and development of glioma by identifying the m6A methylation modification of GAS5 is expected to provide a new method for the diagnosis and treatment of glioblastoma. |
