粉尘螨滴剂对Ⅰ型变态反应的影响

 目的考察粉尘螨滴剂对Ⅰ型变态反应的影响。方法2,5,10倍临床拟使用剂量的粉尘螨滴剂于大鼠或豚鼠舌下给药4～5周,进行大鼠的同种被动皮肤过敏反应(PCA反应)实验、豚鼠支气管哮喘实验、致敏豚鼠离体回肠平滑肌过敏性收缩实验(Schultz Dale test)、大鼠肥大细胞释放组胺实验和致敏大鼠肺中白细胞浸润实验。结果粉尘螨滴剂按拟定的临床给药方案给药4～5周能够明显减小大鼠PCA反应的蓝斑直径和染料的渗出,明显延长卵白蛋白致豚鼠死亡时间,降低豚鼠死亡率,抑制豚鼠回肠Schultz Dale反应时的收缩(P<0.05),抑制抗原再次刺激时大鼠肥大细胞释放组胺,明显减少致敏大鼠抗原再次刺激时肺内过敏性白细胞渗出(P<0.01)。结论粉尘螨滴剂舌下给药4~5周能抑制Ⅰ型变态反应。

Effects of Dermatophagoides farinae Drops on Type Ⅰ Hypersensitivity Reaction

OBJECTIVE To study the effect of Dermatophagoides farinae drops on type Ⅰ hypersensitivity reaction.METHODS Dermatophagoides farinae drops(2,5 and 10 times of the clinical intended dosages)were administered sublingually to Sprague-Dawley rats or guinea pig for 4～5 weeks.Passive cutaneous anaphylaxis(PCA reaction) in rats,bronchia asthma test in guinea pig,Schultz Dale reaction using guinea pig ileum,histamine-releasing test in the mast cell of rats,leukocytes infiltration test in the right lung of sensitized rats were performed.RESULTS After administering Dermatophagoides farinae drops,the diameter of the blue spot and exudation of Evans blue were lessoned,the shrink of ileum in the Schultz Dale reaction in sensitized guinea pigs were decreased(P<0.05),the amount of histamine released from the mast cell in rats and the number of the leukocytes infiltrating to the lung in sensitized rats were reduced,and the death time resulted from another albumen attack was prolonged.CONCLUSION Dermatophagoides farinae drops could inhibit the type Ⅰ hypersensitivity reaction.
