蛋白质缺乏对小鼠腹腔巨噬细胞的影响

摘    要:
采用间接免疫荧光定性试验(IFAT),免疫酶染色试验(IEST)、荧光定量试验、巨噬细胞(Mφ)吞噬功能测定、扫描电镜(SEM)及免疫电镜(IEM)对蛋白质缺乏小鼠腹腔Mφ的免疫反应、超敛结构和吞噬功能进行了观察与研究。结果表明:小鼠在给予缺乏蛋白质饲料后,体重进行性下降。腹腔Mφ膜表面的荧光反应强度、免疫酶染色反应随之减弱;Mφ膜表面所结台的荧光抗体量减少;SEM下可见Mφ表面绒状突起变短;IEM显示细胞阳性反应率及反应程度下降,胞膜及核膜反应减弱;Mφ吞噬功能抑制。上述结果提示蛋白质缺乏对小鼠腹腔Mφ的免疫反应、结构及功能均可产生明显影响。

Abstract:
We have observed and studied the immune response, ultrastructure and phagocytosis of peritoneal macrophages (Mφ) of mice in protein deficiency by means of indirect fluorescent antibody test (IFAT), immunoenzymatic staining technique (IEST),fluorescence isothiocyanate antibody (FITC-Ab) quantitative assay, Mφ phagocytosis test, scanning electron microscopy (SEM) and im-munoelectron microscopy (IEM).The results showed that the body weight of mice was continuously declined after fed protein deficient diet. In the same time fluorescence reaction and enzyme stain on the Mφ surface was retarded. The amount of FITC-Ab on the Mφ membrane was decreased. The villi on the Mφ surface were shortened, the positive rates and positive degree of cells were lowered,the reaction of cell membrane and nuclear membrane was retarded in SEM and IEM.The phagocytic function of Mφ was inhibited.The results showed that in protein deficiency, the immune reaction, structure and function of peritoneal Mφ of mice were markedly affected.
