冬枣提取物对S180荷瘤小鼠的免疫及抗氧化作用

摘    要:目的 观察冬枣提取物(DJE)对S180荷瘤小鼠的免疫及抗氧化作用.方法 取昆明种小鼠50只,先设正常对照组10只, 其余小鼠左腋皮下接种瘤液0.2 mL,24 h 后随机分为肿瘤对照组、冬枣乙醇提取物(EED)组、冬枣乙酸乙酯萃取物(EAED)组,环磷酰胺(CTX)组.正常对照组、肿瘤对照组用生理盐水灌胃, 每天1次;EED组、EAED组分别给予EED 100 mg/(kg·d)、 EAED 100 mg/(kg·d) 灌胃;CTX组给予CTX 20 mg/kg,第2天起隔日腹腔注射1次.实验连续10 d.末次给药24 h后,将小鼠称质量.眼眶取血, 测定各组丙二醛(MDA)含量及超氧化物歧化酶(SOD)活力,采用双抗体夹心ELISA法测定各组血清白细胞介素2浓度.剥取肿瘤、胸腺、脾脏, 计算抑瘤率、胸腺指数、脾指数.结果 DJE可明显抑制S180荷瘤小鼠瘤体生长,肿瘤抑制率最高达到24.8%;DJE干预组胸腺指数、脾指数与肿瘤对照组比较均无明显差异(P>0.05),但均较CTX组显著增高(F=3.912、8.924,q=2.502~4.654;P<0.05);DJE干预组小鼠血清中白细胞介素-2含量及SOD活力均较肿瘤对照组显著增加(F=2.932、41.299,q=2.839~12.222,P<0.05),MDA含量则明显低于肿瘤对照组(F=12.917,q=5.812、3.196,P<0.05).结论 DJE具有抑制S180 实体瘤生长,提高荷瘤小鼠免疫功能以及增强抗氧化防御系统酶活性的作用.

Abstract:Objective To study the effects of Dongzao Jujube Extract (DJE) on immune function and antioxidation in S180-bearing mice.Methods This study consisted of 50 Kunming mice. Ten mice served as normal control (NC) group. The other 40 received subcutaneous inoculation on the left axilla of 0.2 Ml normal saline containing tumor cells, and randomized to four groups as tumor control (TC), Ethanol Extraction of Dongzao Jujube (EED), Ethyl Acetate Extraction of Dongzao Jujube (EAED) and Cyclophosphamide (CTX) group. The mice in the NC and TC groups were given saline solution by gavage once a day; those in EED and EAED groups, 100 mg/kg EED and EAED, respectively; those in CTX group were intraperitoneally injected with 20 mg/kg CTX every other day. The experiment lasted for 10 days. The mice were weighed after 24 hours of the final medication, blood samples collected from eye sockets for detection of malondialdehyde (MDA) and the activity of superoxide dismutase (SOD). Serum interleukin (IL)-2 was determined by sandwich ELISA. The tumor, thymus gland and spleen were collected and oncostatic rate, thymus index and spleen index calculated. Results DJE significantly inhibit the growth of S180 tumor in mice, with the maximal tumor-inhibition rate reaching 24.8%. The differences of thymus and spleen index between DJE and TC groups were not significant (P>0.05), but apparently higher than that in CTX group (F=3.912,8.924;q=2.502-4.654;P<0.05). The serum level of IL-2 and SOD activity in DJE groups were significantly higher than that in the TC (F=2.932,41.299;q=2.839-12.222;P<0.05), but the level of MDA was significantly lower (F=12.917;q=5.812,3.196;P<0.05).Conclusion DJE possesses effects of inhibiting the growth of sarcoma S180, improving the immune function and strengthening antioxidation defence system in tumor-bearing mice.
