缬草醇提物和水提物萃取组分对小鼠的抗抑郁作用

摘    要:
目的:观察缬草醇提物和水提物萃取组分的抗抑郁作用.方法:100只小鼠随机分为10组,每组10只.阴性对照组给予生理盐水20 mL/kg,阳性对照组给予安乐片20 mg/kg,缬草醇提物Ⅰ、Ⅱ、Ⅲ、Ⅳ组分别给予缬草醇提物水相、氯仿相、乙酸乙酯相和正丁醇相(200、5、10和25 mg/kg),缬草水提物Ⅰ、Ⅱ、Ⅲ、Ⅳ组分别给予缬草水提物水相、氯仿相、乙酸乙酯相和正丁醇相(200、5、10和25 mg/kg),灌胃给药,连续10 d.给药第10天,用光电计数法测5 min内小鼠自主活动;给药第11天,记录5 min内小鼠强迫游泳不动时间;给药第12天,记录6 min内小鼠悬尾不动时间.结果:10组小鼠自主活动时间相比差异无统计学意义(F=1.418,P=0.260),强迫游泳及悬尾不动时间相比差异有统计学意义(F=99.670和106.564,P<0.001),且醇提物Ⅲ、Ⅳ组和水提物Ⅲ、Ⅳ组强迫游泳及悬尾不动时间均较阴性对照组缩短(P<0.05).结论:缬草醇提物及缬草水提物的乙酸乙酯相和正丁醇相均有不同程度的抗抑郁作用.

Abstract:
Aim:To observe antidepressive effects of the ethanol-extraction and water-extraction components of Valeriana officinalis L.on mice.Methods:A total of 100 mice were allocated into 10 groups,10 mice in each group.The negative control group was given normal saline at 20 mL/kg,the positive control group was given Anle tablets at 20 mg/kg,the valerian ethanol-extraction componentsⅠ,Ⅱ,Ⅲ,and Ⅳ groups were given valerian ethanol-extraction components in water,chloroform phase,ethyl acetate phase and n-buoh extraction phase at 200,5,10,and 25 mg/kg,and the valerian water-extraction componentsⅠ,Ⅱ,Ⅲ,and Ⅳ groups were given valerian water-extraction components in water,chloroform phase,ethyl acetate and n-buoh extraction phase at 200,5,10,and 25 mg/kg.All the groups were administered by gavage for 10 days.On the 10th day,the photoelectric counting method was used to record the independent activities of mice within 5 min;on the 11th day,the forced swimming time of mice within 5 min was recorded;on the 12th day,the tail suspension fixed time within 6 min was recorded.Results:There was no statistical differences for 10 groups in independent activities time(F=1.418,P=0.260),but there were statistical differences in the forced swimming time and tail suspension fixed time(F=99.670 and 106.564,P<0.001),and compared with the negative control group,the forced-swimming time and tail suspension fixed time of ethanol-extraction components Ⅲ and Ⅳ groups and water extraction components Ⅲ and Ⅳ groups were shorter(P<0.05).Conclusion:Ethyl acetate and n-buoh extraction phases of valerian ethanol-extraction components and water extraction components have antidepressive effects.
