皮肤鳞状细胞癌组织中埃兹蛋白和水通道蛋白3的表达

摘    要:
目的:探讨埃兹蛋白(Ezrin)和水通道蛋白3(AQP3)在皮肤鳞状细胞癌(SCC)组织中的表达及临床意义。方法:应用免疫组化染色SP法检测65例皮肤SCC(患者中男36例,女29例;高分化24例,中分化23例,低分化18例;有颈淋巴结转移9例;均无合并症且未经治疗)和20例正常皮肤组织中Ezrin和AQP3的表达情况。结果:SCC组织中Ezrin、AQP3阳性表达率分别为81.5%(53/65)和86.2%(56/65),2者在正常皮肤组织中的阳性表达率均为5.0%(1/20),SCC组织中Ezrin和AQP3阳性表达率高于正常皮肤组织(χ2分别为38.669,45.598,P均<0.001)。Ezrin的表达与SCC的分化程度有关,分化程度越低阳性表达率越高(P=0.024);且与淋巴结转移无关(χ2校正=0.022,P=0.881)。AQP3蛋白的表达与SCC的分化程度有关,随SCC分化程度的增高而升高(P=0.012);与淋巴结转移无关(χ2校正<0.001,P>0.999)。SCC组织中Ezrin和AQP3的表达呈正关联(rP=0.446,P<0.001)。结论:Ezrin和AQP3的高表达可能参与了皮肤SCC的发生、发展和转移,并与其恶性程度相关。

Abstract:
Aim:To investigate the expressions and the clinical significance of Ezrin and aquaporins-3(AQP3) proteins in skin squamous cell carcinoma(SCC) tissue.Methods:Ezrin and AQP3 proteins were detected in 65 cases of skin SCC(36 males,29 females;24 cases of well differentiated,23 cases of moderately differentiated and 18 cases of poorly differentiated;9 cases with lymph node metastasis;all without complications and treatment) and 20 normal skin specimens by SP immunohistochemical technique.Results:The positive expression rates of Ezrin and AQP3 in SCC were 81.5%(53/65) and 86.2%(56/65),both of them in normal skin tissue was 5.0%.The positive expression rates of Ezrin and AQP3 in SCC were higher than those in normal skin tissue(χ2=38.669,45.598,P<0.001).In SCC,the expression of Ezrin increased from highly differentiated to poorly differentiated tissue(P=0.024),and was not related to lymph node metastasis(χ2adj=0.022,P=0.881).The positive expression rate of AQP3 decreased from highly differentiated to poorly differentiated tissue(P=0.012),and was not related to lymph node metastasis(χ2adj<0.001,P>0.999).The expression of Ezrin was positively associated with that of AQP3 in SCC tissue(rP=0.446,P<0.001).Conclusion:The high expressions of Ezrin and AQP3 proteins may contribute to the pathogenesis and progression and metastasis,and malignant degree of skin SCC.
