摘    要:目的 探讨Na -H 交换泵-1(Na -H exchanger1,NHE-1)反义基因转染对人胃癌细胞内pH值(intracellular pH,pHi)的调节作用及其在肿瘤基因治疗中的价值.方法 构建人NHE-1基因反义真核表达载体,采用脂质体法将其转染至SGC-7901胃癌细胞中, 采用荧光探针检测转染前后细胞内pH值的变化以及对细胞增殖和凋亡的影响.结果 转染反义基因的细胞pHi明显降低,增殖能力降低,细胞凋亡率增加.结论 NHE-1基因的表达在肿瘤细胞pHi及增殖和凋亡调节中起重要作用.

Abstract:Objective To study the effects of the transefection of antisense human Na -H exchanger 1 (NHE-1) on the pHi of SGC-7901 and its potential role in the gene therapy for cancers. Methods Eukaryotic expression vector pcDNA3.1 encoding antisense NHE-1 was constructed with recombinant DNA technique and transfected into SGC-7901 with liposome DOTAP. The pHi changes of the transfected cells and untransfected cells was detected with a fluorescent probe, BCECF. The proliferation and apoptosis of the cells was studied with flow cytometry. Results Antisense NHE-1 gene transfection induced the acidification, decreased proliferation, and elevated apoptosis of SGC-7901 cells. Conclusion The results suggest that NHE-1 gene plays an important role in the regulation of pHi, reproduction and apoptosis of tumor cells. Inhibiting the over-expression and activity of NHE-1 may be of important clinical value.
