PCOS大鼠窦状卵泡卵母细胞超微结构变化

摘    要:目的:探讨多囊卵巢综合征(PCOS)大鼠的窦状卵泡卵母细胞超微结构的变化。方法:建立PCO 大鼠模型,分别取模型组与对照组双侧卵巢组织,光镜下剔除闭锁及囊状卵泡,选择发育正常窦状卵泡卵母细 胞进行透射电镜观察,比较两组窦状卵泡的一般情况,以及卵母细胞中出现异常的例数,并进行统计分析。结 果:正常卵泡发育到窦状卵泡时,卵母细胞体积逐渐达到最大,胞浆中细胞器丰富,透明带均匀,微绒毛由垂直 细长变粗,并逐渐退出透明带。PCOS组多数窦状卵泡卵母细胞胞浆中出现局部细胞器变性、聚集成块,局部胞 浆板层样结构消失,透明带呈不均质状等异常现象。PCOS组卵母细胞异常的发生率显著高于对照组(χ2= 7.2487,P<0.05)。结论:PCOS大鼠窦状卵泡卵母细胞呈现早期细胞凋亡的特征,是卵泡开始退化的标志,也 是卵泡未能发育成熟为优势卵泡的原因之一。

Abstract:Objective:to investigate the ultrastructure of oocyte in antral follicles in rats with polycystic ovarian syndrome (PCOS). Methods: PCOS models were established using rats. Bilateral ovarian tissue was then harvested from both PCOS model and control groups, removed of all atretic and cystic follicles under light microscope. Oocytes of antral follicles were chosen for observation under transmission electron microscope and intergroup comparison of follicular ultrastructure and prevalence of abnormal oocytes. Results: compared with controls, a large majority of oocytes in PCOS models presented with localized organelle degeneration and aggregation, diminishing cytoplasm lamellae and inhomogeneous zona pellucida. Prevalence of abnormal oocytes was also higher in PCOS model group (P<0.05). Conclusion: changes in oocytes in PCOS model were characteristic of early apoptosis, an initial sign of follicle degeneration, and may possibly contribute to their unsuccessful development into dominant follicles.
