摘    要:目的:研究西藏红雪茶多糖含量测定的方法.方法:采用苯酚-硫酸法测定总糖含量,3,5-二硝基水杨酸(DNS)法测定还原糖含量,两者的差值即为多糖含量.结果:总糖标准曲线方程为OD =0.051 4C- 0.0146(r=0.999 2),还原糖标准曲线方程为OD =0.022 9C-0.086(r =0.999 9),两者分别在2.5 ~12.5,8~40 mg·L-1有良好的线性关系.测得LC,LCA,LCP多糖含量分别为63.6%,82.6%,57.6%.结论:该方法操作快速、简便、灵敏、准确、重复性好.

Abstract:Objective: To establish a method for the determination of polysaccharide in Lethariella cladonioides. Method: Using microplate reader, the content of total sugar was determined by means of phenol-sulfuric acid method and 3,5-dinitro salicylic acid (DNS) method was used for the determination of reducing sugar, the difference between the two was the polysaccharide content. Results: The total sugar standard curve equation was OD=0.051 4C-0.014 6(r=0.999 2) and reducing sugar standard curve equation was OD=0.022 9C-0.086 (r=0.999 9). Respectively the linear range was 2.5-12.5 mg·L-1 and 8-40 mg·L-1. The polysaccharide content of LC, LCA, LCP were 63.6%, 82.6%, 57.6%. Conclusion :The method is fast, simple, sensitive, accurate and reproducible.
