急性坏死性胰腺炎肺损伤大鼠高迁移率族蛋白B1的表达及意义

摘    要:目的:观察急性坏死性胰腺炎(ANP)肺损伤大鼠血清高迁移率族蛋白B1(HMGB1)的表达,探讨HMGB1在急性坏死性胰腺炎并发急性肺损伤(ALI)中的意义及机制。方法:将100只SD大鼠随机分成假手术组(SO组)和模型组(ANP组)。各组分别在6h、12h、24h、36h、48h采用酶联免疫吸附法测定血清中HMGB1,同时行动脉血气分析,观察胰腺和肺组织病理形态学的改变。结果:ANP组血清HMGB1从12h开始明显上升,48h达到高峰,与SO组比较,差异有统计学意义(P<0.01)。随着胰腺损伤的逐渐加重,肺损伤程度也在逐渐加重,PaO2进行性下降,PaCO2进行性升高。结论:血清HMGB1的增高与急性坏死性胰腺炎致ALI病情严重程度呈正相关。

Abstract:Objective:To investigate the role and the passible mechanism of HMGB1 in acute necrotizing pancreatitis-associated lung injury in rats.Methods:One hundred Sprague-Dawley rats were randomly divided into two groups: sham operation group(SO group)and acute necrotizing pancreatitis group(ANP group).Animals in each group were sacrificed at 6 h,12 h,24 h,36 h and 48 h af-ter operations respectively.The concentrations of serum HMGB1 were determined by enzyme-linked immunosorbent assay(ELISA).Blood gas analysis was performed.Moreover,pancreatic and pulmonary pathomorphology was compared between the two groups.Re-sults:In the ANP group,serum HMGB1 increased markedly at 6 h,and culminated at 48 h.There was a significant difference in serum HMGB1 concentrations between the ANP group and the SO group(P<0.01).The severity of lung injury was parallel with the severity of pancreas injury.Conclusions:The extent of serum HMGB1 increase is positively correlated with the severity of ALI.
