p21WAF1基因多态性与胃癌的关系

目的探讨p21 WAF1基因缺失与多态性及其在胃癌发生中的意义.方法采用聚合酶链反应-限制性片断长度多态性(PCR-RFLP)法检测胃癌中p21 WAF1基因缺失与多态性.结果 PCR扩增显示, 30例胃癌和45例非胃癌标本均无p21 WAF1第2外显子缺失.PCR-RFLP 发现,胃癌和非胃癌p21 WAF1第2外显子多态性分别为27%(8/30)和9%(4/45), 差异有显著性(χ2=4.23,P<0.05) ,且8例具多态性胃癌标本中的正常胃粘膜亦具有这种多态性.结论胃癌中p21 WAF1基因无缺失,而存在多态性,p21 WAF1基因多态性与胃癌发生有关.

Relationship Between p21 WAF1 Gene Polymorphism and Carcinogenesis in Gastric Carcinoma

Objective:To inverstigate the relationship between p21 WAF1 gene deletion and polymorphism,and role of p21 WAF1 in carcinogenesis of gastric carcinoma .Methods:Deletion and polymorphism of p21 WAF1 gene in exon 2 were examined by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP) analysis. Results: There was no deletion of p21 WAF1 gene in exon 2 in 30 cases of gastric carcinoma and 45 cases of non-gastric carcinoma.However,polymorphism of p21 WAF1 gene in exon 2 was found in 27%(8/30) of gastric carcinoma and 9%(4/45) of non-gastric carcinoma, difference between the two groups was significant(P<0.05).Forthermore, polymorphism of p21 WAF1 gene was also found in normal gastric mucosa of 8 cases of gastric carcinoma with polymorphism.Conclusion: There is p21 WAF1 polymorphism, no p21 WAF1 deletion, in gastric carcinoma.
