摘    要:目的:建立去甲斑螫酸钠冻干粉针含量测定方法。方法:以Fusion—RP80色谱柱(250mm×4.6mm,4μm)为分离柱,以甲醇.KH2PO4缓冲液(0.1mol·L^-1KH2PO4溶液1000mL,加三乙胺1mL,用磷酸调节pH3.0)(15:85)为流动相,流速为1.0mL·min^-1,检测波长为210nm。结果:去甲斑蝥素浓度在2.0~7.0mg·mL^-1。范围内线性关系良好,r=0.9997,平均回收率为100.19%(n=9),RSD为0.15%。结论:该方法适用于去甲斑蝥酸钠冻干粉针的质量控制。

Abstract:Objective: To establish a RP-HPLC method for the determination of sodium demethylcantharidate for injection. Methods: A Fusion - RP80 column (250 mm × 4.6 mm, 4 μm) was used for separation. The mobile phase was methanol-potassium dihydrogen phosphate buffer (adding 1 mL triethylamine to 1 000 mL 0.01 mol · L^-1 potassium dihydrogen phosphate solution and adjusting pH to 3.0 with phosphoric acid)(15 : 85). The flow rate was 1.0 mL · min^-1. The detection wavelength was 210 rim. Results: The standard curve was in good linearity over the range of 2.0 - 7.0 mg · mL^-1 (r = 0.999 7) and the average recovery was 100.19% (n = 9) with RSD 0.15%. Conclusion: The method can be used as the quality control of sodium demethylcantharidate for injection.
