糖尿病大鼠肾脏内皮生长因子及受体的表达意义

目的 探讨血管内皮生长因子（VEGF）及其受体（VEGFR）在糖尿病大鼠肾组织中的表达及其意义。方法 将Wistar雌性大鼠随机分为正常对照组（NC组）和糖尿病模型组（DM组）各16只,于成模后第2,4周各处死8只。采用特异性抗体和免疫组化染色方法,对每例大鼠肾组织中VEGF中VEGFR表达的分布范围及强度变化进行观察,并结合生化指标及肾脏病理进行了分析。结果 糖尿病大鼠肾组织中VEGF及VEGFR表达的分布范围及强度与正常对照组比较,第2周组有升高趋势,第4周组升高呈显著性差异（P＜0．01）,尿白蛋白排泄率（UAER）亦显著升高（P＜0．01）,且肾组织中呈现肾小球内皮细胞增生,典型的结节性变等病理改变。结论 肾局部VEGF及VEGFR表达的增加参与也糖尿病肾病（ND）的发生。

Expression of Kidney Vascular Endothelial Growth Factor and Its Receptor in Diabetic Nephropathy in Rats

Obejective To investigate the clinical meaning of VEGF and VEGFR expression in diabetic nephropathy in rats. Methods Wistar female rats were randomly subdivided into normal control group(NC group) and diabetic model group (DM group), Each group had 16 rats, 8 of each group were killed at 2 weeks and 4 weeks after injection respectively, The change in sites and staining intensity of VEGF and VEGFR expression in renal tissue of each rat was observed by specific antibody (anti -VEGF and anti -VEGFR antibodies) and immuno-histochemistry dye. At the same time, biochemical index and renal pathology were investigated. Results As compared with normal controls, both of the distribution and staining intensity of VEGF and VEGFR expression in the kidney of rats with DM at the end of 2 weeks intended to increase, and at the end of 4 weeks increased significantly (P <0. 01), UAER also increased significantly (P <0. 01), and renal pathology such as glomerular endothelial proliferation and nodular lesions appeared. Conclusion The VEGF and VEGFR expression increase in diabetic nephropathy participated in the occurence of DN.
