Protective Effect of Phloretin on COPD Airway by Inhibiting ERK Activation

Abstract:
Objective: To study the protective effect of phloretin on the airway of COPD by inhibiting the activation of extracellular regulatory protein kinase(ERK). Methods: A total of 36 clean SD adult male rats were randomly divided into control group, COPD group and phloretin intervention group, with 12 rats in each group.Rats in the COPD group and the phloretin intervention group were modeled by cigarette smoking(CS) +lipopolysaccharide(LPS) intratracheal instillation of compound factors. After the establishment of the model, rats in phloretin intervention group received phloretin intervention(phloretin dissolved in 1% DMSO solution, and gavage at 100 mg/kg, once a day for 2 weeks). The rats in the control group and the COPD group were given an equal volume of normal saline intragastrically. The general situation of rats in each group after modeling was compared. The pathological situation of lung tissue in each group was compared by HE staining. The lung function indexes of rats in each group were detected and compared. The levels of serum inflammatory factors(IL-1β,TNF-α) were detected by ELLSA method. The expression of mucin 5AC and ERK protein were detected by Western blotting. The relative expression of mucin 5AC mRNA and IL-1β mRNA was detected by real-time quantitative PCR(RT qPCR). Results: After the establishment of the model, the model rats showed typical symptoms of COPD. The FEV0.3/FVC and dynamic lung compliance of rats in phloretin intervention group were significantly higher than those in COPD group(P<0.05), and airway resistance, IL-1β and TNF-α were significantly lower than those in COPD group(P<0.05). The relative expressions of mucin 5 AC protein, mucin 5 AC mRNA and IL-1β mRNA in lung tissue of rats in phloretin intervention group were significantly lower than those in control group(P<0.05). The expression of P-ERK1/2 in airway epithelium was significantly decreased in phloretin intervention group(P<0.05). Conclusion: phloretin can effectively improve the lung function, reduce the level of inflammatory factors and the secretion of mucin 5AC in COPD rats. The mechanism may be related to the inhibition of P-ERK1/2 expression.
